Parallele Synthese, Charakterisierung und zelluläre Pharmakokinetik von fluoreszenten zell-penetrierenden Peptiden

The cellular delivery of bioactive molecules by cell-penetrating peptides (CPPs) is gaining significance in biomedical research. In contrast to the major part of the investigations published until 2003, recent work has demonstrated that endocytosis plays a major role in the cellular import of several CPPs into mammalian cells. This PhD thesis describes the development of optimized protocols for the reliable synthesis of large collections of fluorescently labelled peptides, especially CPPs, and the application of these molecules for gaining new insights into the intracellular fate of CPPs. The intracellular trafficking and proteolytic breakdown was investigated by a combination of confocal laser scanning microscopy, biophysical techniques, highly sensitive instrumental analytics and pharmacological intervention. The application of carboxyfluorescein in solid-phase synthesis and the establishment of the synthesis of doubly-labeled peptides are presented in the first part. Side products, generated when applying earlier protocols for the in situ activation of carboxyfluorescein, were eliminated by a simple procedure. In order to enable further chemical derivatization of carboxyfluorescein-labeled peptides in solid-phase synthesis, the on-resin introduction of the trityl group was developed as a protecting group strategy for carboxyfluorescein. This protecting group strategy was exploited for the synthesis of peptides labeled with two different fluorescent dyes. Tritylation and optimized labeling conditions led to the development of a fluorescein-preloaded resin for the automated synthesis of fluorescein-labeled compound collections with uniform labeling yields. These protocols were applied to the synthesis of a collection of fluorescently labelled CPPs. The dependence of the import efficiency on the identity of the fluorophore, on the position of the fluorophore in the peptide as well as on the nature of the cargo of the CPPs was addressed. Cellular uptake was quantified by flow cytometry and fluorescence correlation microscopy (FCM). The sequences were based either on the sequence of the Antennapedia homeodomain-derived Penetratin peptide or the Kaposi bFGF-derived MTS-peptide. With respect to the intracellular fate of cationic CPPs, the role of endosomal acidification and retrograde transport for the uptake of the highly basic cell-penetrating peptides Penetratin, Tat and oligo-arginine was investigated. In MC57 fibrosarcoma cells the inhibitors of endosomal acidification chloroquine and bafilomycin A1 abolished the release of the peptides into the cytoplasm. Release into the cytosol preserved endosomal integrity. Using nordihydroguaiaretic acid, a compound that promotes the rapid retrograde movement of both Golgi stack and trans Golgi network membranes back to the endoplasmic reticulum, evidence for an involvement of retrograde transport in the cytoplasmic delivery of cationic CPPs was obtained. A Penetratin analogue terminally labeled with two different fluorophores was synthesized and used as a sensor to quantitatively dissect the contribution of intracellular proteolytic activities on break-down by fluorescence microscopy and spectroscopy. Fluorescence resonance energy transfer was employed as a read-out in fluorescence spectroscopy. Using a panel of protease inhibitors, the endocytic compartment was identified as the major site of degradation. Inhibition of the proteasome had little but significant effect on intracellular peptide integrity. The ability to monitor break-down of these molecules inside the cell will enable a rational optimization of the peptide-based delivery of vaccines and therapeutic oligonucleotides.Zell-penetrierende Peptide (ZPPs) erlangen in der biomedizinschen Forschung zunehmende Bedeutung im zellulären Import bioaktiver Moleküle. Bis zum Jahr 2003 wurde ein nicht-endozytotischer Aufnahmemechanismus für eine ganze Reihe von ZPPs favorisiert. Neuere Arbeiten konnten allerdings zeigen, dass Endozytose an der Internalisierung von einer Reihe von ZPPs beteiligt ist. In der vorliegenden Doktorarbeit wird die Entwicklung von neuen, effizienten Syntheseverfahren für fluoreszent markierte Peptide und die Anwendung dieser Verfahren speziell auf ZPPs beschrieben sowie der Einsatz dieser Moleküle zur Aufschlüsselung des zellulären Verbleibs gezeigt. Der Aufnahmemechanismus und der proteolytische Abbau von ZPPs wurde detailliert durch eine Kombination konfokaler Laserscanningmikroskopie, biophysikalischer Methoden, hochempfindlicher instrumenteller Analytik und pharmakologischer Intervention untersucht. Die Anwendung von Carboxyfluoreszein in der Festphasenpeptidsynthese wird im ersten Teil der Arbeit gezeigt. Durch in situ Aktivierung von Carboxyfluoreszein entstehende Nebenprodukte wurden identifiziert und durch ein einfaches Protokoll entfernt. Die Einführung einer Trityl-Schutzgruppe ermöglichte sowohl die effiziente Synthese von doppelt fluoreszent markierten Peptiden, als auch die Entwicklung eines Synthese-Harzes für die parallele Herstellung von C-terminal markierten Peptiden. Die entwickelten Protokolle wurden auf die Synthese einer Kollektion von fluoreszent markierten ZPPs angewendet. Der Einfluss des Fluorophors, der Position des Fluorophors innerhalb des Peptids und des Cargos auf die Aufnahme von ZPPs wurden untersucht. Die zelluläre Aufnahme wurde mittels Durchflusszytometrie und Fluoreszenzkorrelationsmikroskopie quantifiziert. Die ZPP-Sequenzen basierten entweder auf dem Antennapedia homeodomain-abgeleiteten Penetratin-Peptid oder dem Kaposi bFGF-abgeleiteten MTS-Peptid. Bei der Untersuchung des intrazellulären Weges konnte gezeigt werden, dass die endosomale Ansäuerung ein Schlüsselereignis für die Freisetzung kationischer ZPPs ins Zytoplasma darstellt. Für die hochbasischen ZPPs Penetratin, Tat und Oligoarginin verhinderten die beiden Inhibitoren der endosomalen Ansäuerung Bafilomycin A1 und Chloroquin die Freisetzung ins Zytoplasma. Die Freisetzung der kationischen ZPPs ins Zytosol beeinträchtigte nicht die endosomale Integrität. Durch Einsatz von Nordihydroguaiarinsäure, ein Wirkstoff, der den Transfer von Membranen des Golgi-Komplexes und Trans-Golgi-Netzwerkes zum Endoplasmatischen Retikulum fördert, wurden Hinweise auf eine Beteiligung des retrograden Transportes am intrazellulären Weg von ZPPs gewonnen. Im letzten Teil der vorliegenden Arbeit wurde ein Penetratin-Analogon mit zwei verschiedenen Fluorophoren synthetisiert. Dieses ZPP ermöglichte die Untersuchung des intrazellulären Abbaus des Peptides über Fluoreszenz-Resonanz-Energietransfer mittels einer Kombination von Fluoreszenzmikroskopie und Fluoreszenzspektroskopie. Durch Einsatz einer Reihe von Protease Inhibitoren konnte gezeigt werden, dass das endosomale Kompartiment der Hauptort der intrazellulären Degradation diese Penetratin-Analogons darstellt, wohingegen das Proteasom wenig zum Abbau beiträgt. Die Fähigkeit, den intrazellulären Abbau von ZPPs zu verfolgen, wird einen rationalen Zugang zum effizienteren peptid-vermittelten Import von Vakzinen und Oligonucleotid-basierten Wirkstoffen ermöglichen
