The cellular delivery of bioactive molecules by cell-penetrating peptides (CPPs) is gaining significance in biomedical research. In contrast to the major part of the investigations published until 2003, recent work has demonstrated that endocytosis plays a major role in the cellular import of several CPPs into mammalian cells. This PhD thesis describes the development of optimized protocols for the reliable synthesis of large collections of fluorescently labelled peptides, especially CPPs, and the application of these molecules for gaining new insights into the intracellular fate of CPPs.
