Conformationally constrained analogues of natively occuring bioactive polypeptides are valuable tools for the understanding of their structure-function-relationships and for the design and development of lead structures with therapeutic and diagnostic applications. The work described in this thesis focuses on the application of conformational constraints on two polypeptides from the calcitonin gene peptide "superfamily": the calcium- and bone-turnover-regulating hormone calcitonin (Ct) and the carbohydrate metabolism-regulating hormone islet amyloid polypeptide (IAPP).
