In der vorliegenden Doktorarbeit wurden die strukturellen und funktionellen Eigenschaften des Parkinson-assoziierten Proteins DJ-1 anhand seiner Reprimierung des ASK1-JNK/p38-Signalwegs untersucht. Es wurde von uns bereits früher gezeigt, dass DJ-1 ASK1 bindet und seine Aktivität reprimiert. Im ersten Teil der Arbeit wurde untersucht, welche oxidierbaren Aminosäurereste von DJ-1 für seine Aktivierung und die Bindung an ASK1 notwendig sind. Es wurde gezeigt, dass Cystein-106 der wichtigste oxidierbare Rest und notwendig für die Bindung an ASK1 und die cytoprotektive Funktion von DJ-1 ist.
