Identification and characterisation of ependyma- specific genes and their proteins

The ependyma is a single-layered epithelium lining the ventricles of the brain and the central canal of the spinal cord. Its prominent cell type are the ependymocytes that bear a bundle of kinocilia at their apical surface projecting into the cerebrospinal fluid (CSF). Despite ependymal cells were already described in 1836 their specific roles in health and disease are not yet precisely understood. An approach elucidating ependymal functions is the identification of genes that are selectively transcribed in the ependyma but not in the remaining brain parenchyma. Therefore, suppression subtractive hybridisation was used to construct an ependymal cDNA library of “ependyma minus brain parenchyma” and “subcommissural organ minus brain parenchyma”, respectively. Differential screening of both libraries revealed the wdr16 gene to be specifically transcribed in ependymal cells. Wdr16 is a WD40-repeat protein with two predicted covalently linked 7-bladed beta-propellers. Wdr16 is evolutionary conserved and restricted to kinocilia-possessing organisms. The rat ortholog is highly expressed in testis and ependymal primary culture as shown by Western blot analysis. RT-PCR studies demonstrate abundant transcription of the rat wdr16 gene in testis and ependymal primary culture. Moderate Wdr16 mRNA levels are present in lung and brain, whilst kinocilia-free tissues are completely devoid of the Wdr16 message. In testis as well as in ependymal primary culture, levels for wdr16 transcript increase at approximately the same time and with practically identical time courses as the mRNA for the central apparatus constituent sperm-associated antigen 6 and the protein restricted to ciliated cells and associated with hydrocephalus, hydin. Double immunostainings demonstrated that the expression of Wdr16 is tied to the formation of kinocilia in an ependymal primary culture system as well as in testis and the respiratory epithelium. Beyond that, in-situ hybridisation experiments in zebrafish indicate that the gene orthologous to wdr16 is only transcribed in kinocilia-possessing cells. Thus, the wdr16 transcription profile in conjunction with its expression pattern on the protein level implies Wdr16 as a differentiation marker for kinocilia-bearing cells. In order to analyse the role Wdr16 might play in physiological and pathological processes, gene kockdown experiments were conducted. Blocking the translation of Wdr16 mRNA in zebrafish embryos via antisense morpholinos results in the generation of hydrocephalus, while ependymal morphology and ependymal ciliary beat remain unaltered compared to control and wildtype animals. Thus, neither a denudation of the ependymal layer nor defunct kinocilia can be considered as the cause for the observed ventricular dilation. Rather, the situation seems to be similar to the one in the hyh mutant mouse, in which a disturbed cell polarity is the cause for ventricular enlargement. Wdr16 might be an adapter protein that is responsible for the proper targeting of ion channels and transporters to their correct destination. Disturbances of this process are likely to cause altered cerebral ion and water flux, resulting in the generation of hydrocephalus.Das Ependym ist ein einschichtiges Epithel, das die Hirnventrikel und den Zentralkanal des Rückenmarks auskleidet. Der hauptsächlich vorherrschende Zelltyp im Ependym sind die Ependymocyten, die ein Kinozilienbündel auf ihrer apikalen Oberfläche tragen, das in die Cerebrospinalflüssigkeit hineinragt. Obwohl die Ependymzellen seit 1836 beschrieben sind, ist ihre spezifische Funktion in physiologischen und pathologischen Prozessen bis heute nur unzureichend verstanden. Ein Ansatz zur Aufklärung ependymaler Funktionen ist die Suche nach Genen, die spezifisch im Ependym transkribiert werden, nicht jedoch im restlichen Gehirnparenchym. Zu diesem Zweck wurden durch "suppression subtractive hybridisation“ cDNA Bibliotheken generiert, nämlich "Ependym minus Gehirnparenchym“ und "Subcommissuralorgan minus Gehirnparenchym“. Ein differentielles "Screening“ beider Bibliotheken ergab, dass das Wdr16-Gen spezifisch im Ependym transkribiert wird. Wdr16 ist ein Protein mit WD40- Sequenzwiederholungen ("WD40-repeats“) und zwei kovalent verknüpften beta-Propellern. Wdr16 ist evolutionär konserviert und auf Kinozilien tragende Organismen beschränkt. Das Rattenorthologe wird stark im Hoden und in ependymreicher Primärkultur exprimiert, was durch "Western Blot“ Analysen gezeigt werden konnte. RT-PCR-Studien demonstrieren eine häufige Transkription des Ratten Wdr16 Gen, moderate Wdr16-mRNA-Spiegel in Lunge und im Gehirn, während Gewebe ohne Kinozilien keine Wdr16-mRNA aufweisen. Sowohl im Hoden als auch in ependymreicher Primärkultur steigt der Spiegel des Wdr16-Transkripts zum nahezu gleichen Zeitpunkt und mit praktisch identischem zeitlichen Verlauf wie die mRNA des im Axonem-Zentralapparat ansässigen Proteins "Spermien assoziiertes Antigen 6“ und diejenige des Proteins Hydin, dessen Expression auf ziliierte Zelle beschränkt ist und das im Zusammenhang mit Hydrocephalus steht. Doppelimmunfärbungen belegten, dass die wdr16-Expression in ependymreicher Primärkultur, im Hoden und auch im respiratorischen Epithel an die Ausbildung von Kinozilien geknüpft ist. Darüber hinaus zeigen in-situ Hybridisierungsexperimente im Zebrafisch, dass das zu wdr16 orthologe Gen nur in Zellen transkribiert wird, die Kinozilien besitzen. Demzufolge impliziert das wdr16-Transkriptionsprofil in Verbindung mit dem Expressionsmuster, dass es sich bei Wdr16 um einen Differenzierungsmarker für Kinozilien tragende Zellen handelt. Um zu untersuchen welche Rolle Wdr16 in physiologischen und pathologischen Prozessen spielen könnte wurden Gen- "Knockdown“-Experimente durchgeführt. Eine Inhibition der Wdr16-mRNA- Translation in Zebrafischembryonen mittels "antisense“-Morpholinos führt zur Ausbildung von Hydrocephalus, wobei die ependymale Morphologie und der ependymale Zilienschlag im Vergleich zur Kontrolle und zu Wildtyp-Tieren unverändert sind. Demzufolge können weder eine Ablösung der Ependymschicht noch Defekte der Kinozilien als Ursache für die Ventrikeldilatation in Betracht gezogen werden. Vielmehr scheint die Situation ähnlich zu sein wie in der hyh-Mausmutante, wo eine Störung der Zellpolarität ursächlich für die Ventrikelvergrößerung ist. Bei Wdr16 könnte es sich um ein Adapterprotein handeln, dass für den Transport von Ionenkanälen und Transportern an ihren Bestimmungsort verantwortlich ist. Eine Störung dieses Prozesses dürfte einen veränderten Ionen- und Wasserflux nach sich ziehen, was schließlich zur Ausbildung des Hydrocephalus führt
