The fast-pace development of genome-editing technologies brings hope to provide a cure for genetic diseases fundamentally. However, the inefficient delivery of genome-editing systems including zinc-finger nucleases (ZFNs), transcription activator-like effector nucleases (TALENs), and clustered regularly interspaced short palindromic repeats-associated nuclease Cas9 (CRISPR/Cas9) still limits their clinical applications. Compared to systemic delivery, pulmonary delivery enables noninvasive and direct deposition of drug in lung, which has made pulmonary delivery promising for the treatment of many lung diseases including cystic fibrosis (CF).
