Otimização de reatores descontínuos sequenciais de biofilme: resposta a microrganismos filamentosos e compostos antibacterianos

The present work aimed to give additional contributions to solve the problem of filamentous bulking in sequencing batch reactors (SBRs) through the addition of biofilm carriers as well as, to provide useful data for the application of biofilms in SBRs operated in the presence of antibiotics. The influence of the organic loading rate (OLR) on the overgrowth of filamentous organisms was assessed by operating three SBRs fed with different acetate loading rates: 4.3 ± 0.3 g COD L-1 day-1 (control unit), 1.0 ± 0.1 g COD L-1 day-1 and 0.5 ± 0.1 g COD L-1 day-1. The microscopic observations showed that the excessive growth of filaments only occurred in the SBR operated with the highest OLR. Acetate concentration profiles indicated that acetate removal occurred during the entire SBR cycle in the reactor facing the overgrowth of filaments, while in the others two SBRs a period of high substrate concentration followed by a period without substrate (“feast-famine” conditions) was happening. The nonexistence of “feast-famine” conditions in the SBR operated with the highest OLR probably favoured the excessive growth of filaments. The filaments were identified by culture-dependent techniques together with molecular methods as closely related to Geotrichum candidum (fungus). The study of the influence of carrier concentration in the overgrowth of the G. candidum-like filaments and in the bacterial community was conducted in SBRs without (control unit similar to the one of the previous study) and with 5 %, 10 % and 20 % carriers. The occurrence of the fungal filaments was only negligible in the SBRs with 10 % and 20 % carriers, apparently because the higher amount of sludge obtained in these reactors (suspended and biofilm) decreased the substrate availability and the carrier-to-carrier collisions established increased the shear stress. Bacterial community fingerprinting using molecular techniques showed that the bacterial groups usually present in activated sludge systems prevailed. The influence of the wastewater composition in the overabundance of the fungal filaments was also evaluated. Two SBRs were operated: the control SBR was fed with acetate (soluble organic matter) and the other SBR received wastewater containing both soluble and particulate organic matter. The results indicated that the wastewater composition influenced the overgrowth of the G. candidum-like filaments since they were detected in the SBR fed with acetate, while in the other SBR, filamentous bulking caused by Meganema perideroedesrelated filaments was identified (using culture-independent techniques). Both bulking were successfully overcome through the addition of carriers into the SBRs, suggesting that this might be an effective strategy to solve filamentous bulking. The addition of 10 % carriers solved the G. candidum-like bulking, apparently because the combined effect of decreasing the substrate availability and increasing the shear stress induced the growth of the fungus as arthroconidia instead of filaments. The suppression of the M. perideroedes-like bulking was probably related to the increase of the sludge concentration caused by the addition of 20 % carriers. The final study of this dissertation compared the microbial community composition and activity as well as the antibiotic resistance genes of SBRs operated with biofilms and receiving wastewater without and with 50 μg L-1 of tetracycline. The removal of organic carbon and nitrogen were similar in both reactors, but the bacterial composition of the biofilms (identified using culture-independent methods) differed. The presence of tetracycline decreased the fraction of Flavobacterium, Caulobacter and Zooglea-like bacteria and of members of the Sphingobacteriaceae family and increased the fraction of the Sandarakinorhabdus genus. The analysis of the tetracycline resistance (tet) genes indicated a relationship between the occurrence of tet(S) and tetracycline contamination. The occurrence of the others tested tet genes [tet(A), tet(C), tet(G), tet(M) and tet(X)] was not changed by the presence of the antibiotic. The reactor with tetracycline showed poor tetracycline removal (about 28 %): 15.8 ± 0.3 μg of the antibiotic was found sorbed to the biofilm matrix and, based on a mass balance, the remaining antibiotic removal probably occurred by metabolic processes.A proliferação de microrganismos filamentosos e a presença de micropoluentes antibacterianos emergentes são problemas com os quais uma Estação de Tratamento de Águas Residuais se pode confrontar. Este trabalho pretendeu contribuir para a resolução do problema do crescimento excessivo de filamentos em reatores sequenciais descontínuos (SBR, do inglês Sequencing Batch Reactors) através da adição de suportes para a formação de biofilmes assim como, fornecer dados úteis para a aplicação de biofilmes a SBR operados na presença de antibióticos. A influência da carga orgânica no crescimento excessivo de microrganismos filamentosos foi avaliada através da operação de três SBR alimentados com diferentes cargas de acetato: 4.3 ± 0.3 g CQO L-1 dia-1 (reator controlo), 1.0 ± 0.1 g CQO L-1 dia-1 e 0.5 ± 0.1 g CQO L-1 dia-1. As observações microscópicas demonstraram que a proliferação de filamentos apenas ocorreu no reator operado com a carga orgânica mais elevada. Os perfis de concentração de acetato indicaram que a remoção deste ocorria durante todo o ciclo do SBR no reator com problemas de crescimento excessivo de filamentos, enquanto nos outros dois reatores, um período de elevada concentração de acetato seguido por um período de ausência de acetato sucedia. A não existência de condições de abundância e escassez no SBR operado com a carga orgânica mais elevada provavelmente favoreceu a proliferação de filamentos. Estes filamentos foram identificados por técnicas de cultivo em conjunto com técnicas de biologia molecular como intimamente ligados com o fungo Geotrichum candidum. O estudo da influência da concentração de suportes na proliferação dos filamentos fúngicos e na comunidade bacteriana foi conduzido em SBR operados sem (reator controlo similar ao do estudo anterior) e com 5 %, 10 % e 20 % de suportes. A ocorrência dos filamentos fúngicos só foi desprezável nos SBR com 10 % e 20 % de suportes, aparentemente porque a maior concentração de lamas obtidas nestes dois reatores (suspensão + biofilme) fez decrescer a disponibilidade de substrato e as colisões entre suportes que se estabeleceram fizeram aumentar as forças de atrito. O estudo da comunidade bacteriana, através de técnicas moleculares, demonstrou que a presença de suportes induziu alterações na mesma mas, os grupos bacterianos usualmente presentes em sistemas de lamas ativadas prevaleceram. A influência da composição da água residual no crescimento excessivo dos filamentos fúngicos também foi avaliada. Dois SBR foram operados: o SBR controlo foi alimentado com acetato (solúvel) e o outro SBR recebeu matéria orgânica solúvel e particulada. Os resultados indicaram que a composição da água residual influenciou a proliferação dos filamentos fúngicos uma vez que, eles foram detetados no SBR operado com acetato, enquanto no outro reator foi detetado o crescimento excessivo de filamentos afiliados ao Meganema perideroedes (usando técnicas moleculares). Em ambos os casos, a adição de suportes aos SBR suprimiu o crescimento excessivo de filamentos. A adição de 10 % de suportes resolveu o problema da proliferação de filamentos intimamente associados ao fungo G. candidum, aparentemente porque o decréscimo da disponibilidade de acetato e o aumento das forças de atrito induziram o crescimento do fungo como artroconídios em vez de filamentos. A supressão dos filamentos afiliados ao M. perideroedes esteve provavelmente relacionada com o aumento da concentração das lamas causado pela adição de 20 % de suportes. O último estudo desta dissertação comparou a atividade e composição da comunidade microbiana assim como, os genes de resistência a antibióticos de SBR operados com biofilme e recebendo água residual sem e com 50 μg L-1 de tetraciclina. A remoção de matéria orgânica e nitrogénio foi similar em ambos os reatores, mas a composição da comunidade bacteriana dos biofilmes divergiu. A presença de tetraciclina diminui a fração de bactérias relacionadas com os gêneros Flavobacterium, Caulobacter e Zooglea-like bacteria e com membros da família Sphingobacteriaceae e aumentou a fração do gênero Sandarakinorhabdus. A análise dos genes de resistência à tetraciclina (tet) indicou uma relação entre a ocorrência do gene tetS e a presença do antibiótico. A ocorrência dos genes tet(A), tet(C), tet(G), tet(M) and tet(X) não foi alterada pela presença de tetraciclina. O reator com tetraciclina apresentou uma remoção do antibiótico de cerca de 28 %: 15.8 ± 0.3 μg de tetraciclina foram encontradas sorvidas à matriz do biofilme, sendo que a restante remoção do antibiótico ocorreu provavelmente por processos metabólicos
