Novel insights in the neurobiology of depression: a role for neuroplasticity

Nevertheless, the precise physiopathological mechanisms involved in its aetiology and in the therapeutic actions of antidepressant drugs are still largely unknown. Previous studies have implicated different neuroplastic phenomena in the neurobiological basis of depression. In particular, the role of adult neurogenesis in the hippocampus and dendritic and synaptic plasticity in different brain regions in the behavioural actions of antidepressant drugs has been the focus of recent attention in this field of research. In the present work, we addressed the behavioural interactions of mood, anxiety and cognition in an animal model of depression, taking into account the neuroplastic events that might underlie the behavioural actions of different antidepressant drugs. To address the interaction between mood, anxiety and cognition, thorough behavioural assessments were performed after exposure to an unpredictable chronic mild stress (CMS) model of depression. To clarify the role of hippocampal adult neurogenesis in the behavioural actions of antidepressants, different antidepressant drugs were administered with the concomitant ablation of neurogenesis with the cytostatic agent methylazoxymethanol (MAM). Furthermore, dendritic and synaptic plasticity in the hippocampus and prefrontal cortex (PFC) were evaluated by 3-dimensional morphometric analysis and expression of genes encoding for the neural cell adhesion molecule (NCAM) and synaptic protein synapsin1 (SYN1). Finally, these stress-induced neuroplastic phenomena were characterized in the nucleus accumbens (NAc), a brain structure implicated in the core symptom of anhedonia. The results of the present study reveal that the behavioural changes in mood, anxiety and cognition are highly inter-dependent and act in synergistic modes, indicating a continuum between emotional changes and cognitive impairments induced by CMS. Importantly, we concluded that adult neurogenesis in the hippocampus is not required for the mood-improving actions of antidepressants but is associated with the anxiolytic effects of these drugs. In addition, exposure to CMS induces dendritic atrophy and synaptic loss in the hippocampus and PFC which can be reverted with antidepressants and are associated with the altered expression of genes encoding for NCAM and SYN1. Finally, the neuroplastic responses to chronic stress in the NAc associated with anhedonia are characterized by dendritic hypertrophy and gain in synaptic contacts that are normalized by chronic antidepressant treatment and are paralleled with variations in the expression of genes encoding for the brain-derived neurotrophic factor (BDNF), NCAM and SYN1. In conclusion, by extensively characterizing the interactions between the behavioural dimensions involved in depression, the present work has clarified the role of adult neurogenesis in the behavioural actions of antidepressants and has demonstrated the reversibility of stress-induced neuroplastic changes by antidepressants in brain regions associated with learning and memory impairments in depression and in the brain reward pathways associated with anhedonia.A depressão é uma perturbação do humor altamente prevalente que inflige graves consequências nos doentes afectados e um impacto social e económico significativo em todo o mundo. No entanto, os mecanismos fisiopatológicos precisos envolvidos na sua etiologia e nas acções terapêuticas dos fármacos antidepressivos são ainda pouco conhecidos. Estudos anteriores implicaram diversos fenómenos neuroplásticos nas bases neurobiológicas da depressão. Em particular, o papel da neurogénese hipocampal no adulto e da plasticidade sináptica e dendrítica em diferentes regiões cerebrais nos efeitos comportamentais dos antidepressivos tem sido foco de recente atenção nesta área de investigação. No presente trabalho, abordamos as interações comportamentais entre o humor, ansiedade e cognição num modelo animal de depressão, tendo em consideração os eventos neuroplásticos relacionados com os efeitos comportamentais de diferentes fármacos antidepressivos. Para abordar as interacções entre humor, ansiedade e cognição, foram realizadas extensas avaliações comportamentais após exposição a um modelo de depressão de stress crónico moderado (SCM). Para clarificar o papel da neurogénese hipocampal no adulto nos efeitos comportamentais dos antidepressivos, diferentes fármacos antidepressivos foram administrados com ablação concomitante da neurogénese com o agente citostático methylazoxymethanol (MAM). Adicionalmente, a plasticidade dendrítica e sináptica no hipocampo e no córtex pré-frontal (CPF) foram avaliadas por análise tri-dimensional morfométrica e pela expressão dos genes que codificam a molécula de adesão celular neuronal (MACN) e a proteína sinática sinapsina1 (SIN1). Finalmente, estes fenómenos neuroplásticos foram caracterizados no núcleo accumbens (NAc), uma estrutura cerebral implicada no sintoma depressivo de anedonia. Os resultados do presente estudo revelam que as alterações comportamentais no humor, ansiedade e cognição são altamente interdependentes e actuam de uma forma sinérgica, sugerindo um continuum entre as alterações emocionais e os déficits cognitivos induzidos pelo SCM. De salientar, concluímos ainda que a neurogénese hipocampal no adulto não é necessária para os efeitos dos fármacos antidepressivos no humor mas está associada com os seus efeitos ansiolíticos. Adicionalmente, a exposição ao SCM induz atrofia dendrítica e perda sináptica no hipocampo e no CPF que podem ser revertidas com antidepressivos e estão associadas a alterações na expressão dos genes que codificam a MACN e SIN1. Finalmente, as respostas neuroplásticas no NAc associadas com a anedonia são caracterizadas pela hipertrofia dendrítica e aumento de contactos sinápticos, são normalizadas com tratamento antidepressivo e estão associadas a alterações na expressão dos genes que codificam o factor neurotrófico derivado do cérebro (FNDC), MACN e SIN1. Em conclusão, ao caracterizar extensivamente as interacções entre as dimensões comportamentais envolvidas na depressão, o presente estudo clarificou o papel da neurogénese hipocampal no adulto nas acções comportamentais dos antidepressivos e demostrou a reversibilidade das alterações neuroplásticas induzidas pelo stress crónico em regióes cerebrais associadas a déficits de memória e apredizagem e nas vias cerebrais implicadas na anedonia
