Comparación de las técnicas qPCR vs kits rápidos en el Diagnóstico de Leucemia Felina (ViLeF) en el Centro de Diagnóstico Veterinario VETNAAT en la ciudad de Quito.

Feline leukemia is one of the main viral diseases affecting domestic and/or wild felines due to the fact that it has several forms of transmission, among which we find: oral, nasal, sexual, transplacental and intrauterine. For its diagnosis different tests are used such as: ELISA or rapid kits, real-time PCR, Rt-qPCR, immunoassay, immucromatography, among others; of which the most used at clinical level is the ELISA test or rapid kits. For this reason, the objective of the current research project is to compare the sensitivity and specificity of the rapid kits vs. the qPCR molecular test, as well as to analyze the prevalence of risk factors. For this purpose, 26 felines positive or suspicious for Feline Leukemia were used; of which 16 were females and 10 males of different ages and reproductive status; blood samples were collected using the cephalic vein for the rapid kits, and blood plasma for the qPCR molecular test from which the viral RNA was extracted and amplified at the VetNAAT Veterinary Diagnostic Center. For the results, 4 risk factors were taken into consideration, the most important of which are the sex risk factor, with a prevalence of 11.54% for infected males and 15.38% for infected females, and the vaccination status risk factor, with a prevalence of 8.55% for unvaccinated felines and 17.31% for vaccinated felines. According to the data obtained, the disease has no predilection for sex or vaccination status of the felines; qPCR is the most accurate diagnostic technique for the detection of Feline Leukemia (ViLeF), and its results are more reliable. The rapid tests normally used for the diagnosis of the disease in this study have shown low sensitivity.La Leucemia Felina es una de las principales enfermedades de tipo viral que afecta a felinos domésticos y/o salvajes debido a que tiene varias formas de transmisión, entre las cuales encontramos: oral, nasal, sexual, transplacentaria e intrauterina. Para su diagnóstico se utilizan diferentes pruebas como son: ELISA o Kits rápidos, PCR en tiempo real, Rt-qPCR, inmunoensayo, inmucromatografía, entre otras; de las cuales la más utilizada a nivel clínico es la prueba de ELISA o Kits rápidos. Por este motivo, el objetivo del proyecto de investigación en curso consiste en comparar la sensibilidad y especificidad de los kits rápidos vs la prueba molecular qPCR, así como también, analizar la prevalencia de los factores de riesgo. Para lo cual se utilizaron 26 felinos positivos o sospechosos a Leucemia Felina; de los cuales 16 eran hembras y 10 machos de diferentes edades y estado reproductivo; se recolectaron muestras de sangre utilizando la vena cefálica, para los kits rápidos, y plasma sanguíneo para la prueba molecular qPCR del cual se extrajo y amplificó el ARN viral en el Centro Diagnóstico Veterinario VetNAAT. Para los resultados se tomaron a consideración 4 factores de riesgo, de los cuales los más importantes son el factor de riesgo sexo cuya prevalencia fue del 11,54% para machos infectados y 15,38% de hembras infectados y por factor de riesgo por estado de vacunación, tuvo una prevalencia del 8,55% para felinos no vacunados y 17,31% para felinos vacunados. En virtud a los datos obtenidos, la enfermedad no tiene predilección por el sexo o el estado de vacunación de los felinos; la qPCR es la técnica de diagnóstico precisa para la detección de Leucemia Felina (ViLeF), sus resultados son de mayor confiabilidad.
