Aislamiento y estudio de la resistencia a los antibacterianos en cepas de Escherichia coli en aislados de deyecciones en pollos broiler de los criaderos de Poaló del cantón Latacunga.

The study of the sensitivity of bacteria to antibiotics has become an important tool in recent years; it is essential to determine their resistance and evaluate the effectiveness of the treatments that will be used to combat them. This research project assumed the objective of analyzing the resistance to antibacterial (AMR) of the bacterium Escherichia Coli from broiler chickens from Poaló hatcheries. For the research work 50 samples were collected in total, of which 40 were sewage and 10 were taken directly from the droppings present in the soil, were marked and preserved in Stuart means of transport, the ISO 16654: 2001 method was used, in the enrichment process soybean broth with Trypticasein (TSB) was used in test tubes, was inoculated, homogenized and incubated, for selective isolation, MacConkey agar was used with Sorbitol, this was inoculated with the samples previously enriched with the techniques of aseptic transfer and culture in stretch marks, in the process of purification of colonies the samples that met the coloration requirements were selected which is a characteristic rose, caused by the fermentation of lactose, which is associated with enterobacteria, of 50 samples 11 belonging to chickens and 5 floor met the requirements and nutritional agar was used, in this the same planting techniques were used as in the previous agar to inoculate it, with the colonies obtained the Gram staining procedure was done, through the observation with microscope the presence of gram-negative bacteria was confirmed, Microgen biochemical tests were performed, and its Microgen ID software revealed that 5 of the samples from chickens and 2 from the floor had the bacterium Escherichia coli, with these samples the sensitivity tests could be performed, with the use of Mueller Hinton agar and the Bauer & Kirby technique. The results obtained showed colony growth in the inhibition halos, being an indication of resistant populations of E. coli originating from the samples studied: Amoxicillin 100% (resistant), Cephalexin 57% (resistant), 43% (intermediate), Ciprofloxacin 43% (resistant), 14% (Intermediate), 43% (Sensitive), Enrofloxacin 14% (resistant), 72% (intermediate), 14% (sensitive), Florfenicol 86% (resistant), 14% (Sensitive), Fosfomycin 57% (resistant), 14% (intermediate), 29 % (sensitive), Gentamicin 43 % resistant 28% (intermediate), 29% (sensitive), Norfloxacin 29% (resistant) 71% (Intermediate), Penicillin G 100% (resistant) Sulf+Trimetroprim 86 % (resistant), 14% (intermediate),Tetracycline 100% (resistant).El estudio de la sensibilidad que poseen las bacterias hacia los antibióticos se ha convertido en una herramienta importante en los últimos años, resulta esencial para la determinación de su resistencia y evaluar la efectividad de los tratamientos que se usarán para combatirlas. El presente proyecto de investigación asumió el objetivo de analizar la resistencia hacia los antibacterianos (RAM) de la bacteria Escherichia Coli procedente de los pollos broiler de los criaderos de Poaló. Para el trabajo de investigación se recolectaron 50 muestras en total, de las cuales 40 fueron cloacales y 10 se tomaron directamente de las deyecciones presentes en el suelo, fueron marcadas y conservadas en medios de transporte Stuart, se usó el método ISO 16654: 2001, en el proceso de enriquecimiento se usó caldo de soja con Tripticaseína (TSB) en tubos de ensayo, se inoculo, homogenizó y fueron incubadas, para el aislamiento selectivo, se utilizó agar MacConkey con Sorbitol, este fue inoculado con las muestras previamente enriquecidas con las técnicas de transferencia aséptica y cultivo en estrías, en el proceso de purificación de colonias se seleccionaron las muestras que cumplían los requisitos de coloración el cuál es un rosa característico, provocado por la de fermentación de lactosa, lo que se asocia a enterobacterias, de 50 muestras 11 pertenecientes a pollos y 5 de piso cumplieron los requisitos y se empleó agar nutritivo, en este se usaron las mismas técnicas de siembra que en el agar anterior para inocularlo, con las colonias obtenidas se hizo el procedimiento de tinción Gram, a través de la observación con microscopio se confirmó la presencia de bacterias gram negativas, se realizaron las pruebas bioquímicas de Microgen, y su software Microgen ID reveló que 5 de las muestras provenientes de pollos y 2 de piso poseían la bacteria Escherichia coli, con estas muestras se pudo realizar las pruebas de sensibilidad, con la utilización de agar Mueller Hinton y la técnica de Bauer & Kirby. Los resultados obtenidos mostraron crecimiento de colonias en los halos de inhibición, siendo un indicativo de poblaciones resistentes de E. coli originarias de las muestras estudiadas: Amoxicilina 100% (resistente), Cefalexina 57% (resistente), 43% (intermedio), Ciprofloxacina 43% (resistente), 14% (Intermedio), 43% (Sensible), Enrofloxacina 14% (resistente), 72% (intermedio), 14% (sensible), Florfenicol 86% (resistente), 14% (Sensible), Fosfomicina 57% (resistente), 14% (intermedio), 29 % (sensible), Gentamicina 43 % resistente 28% (intermedio), 29% (sensible), Norfloxacina 29% (resistente) 71% (Intermedio), Penicilina G 100% (resistente) Sulf+Trimetroprim 86 % (resistente), 14% (intermedio),Tetraciclina 100% (resistente)
