Effects of mass rearing on the olfactory abilities of an invasive fruit moth species, Grapholita molesta (Busck)

Der Pfirsichwickler, Grapholita (Cydia) molesta Busck (Lepidoptera: Tortricidae), ist ein wirtschaftlich bedeutender Schädling im Stein- und Kernobstbau weltweit. Die Anziehung von befruchteten G. molesta Weibchen zu ihren Wirtspflanzen ist primär über olfaktorische Signale gesteuert. Langjährige Zucht dieser Insekten im Labor kann möglicherweise die olfaktorische Reaktionsfähigkeit verändern. Hier wurden die Reaktionen von Langzeitlaborzuchtpopulationen - 70-80 Generationen (‘2007 Population) - auf die olfaktorische Reaktionsfähigkeit von G. molesta Weibchen auf pflanzliche volatile Stoffe untersucht und mit einer jungen Laborzuchtpopulation - 15 Generationen (‘2009 Population) - verglichen. Anhand eines full-sib/half-sib Versuchsansatzes wurde weiters untersucht, ob die olfaktorische Reaktionsfähigkeit dieser Art eine genetische Komponente aufweist. Ergebnisse zeigten, dass die Laborzucht die olfaktorischen Reaktionsfähigkeiten deutlich beeinflusst und verändert. Weibchen der ‘2007 Population waren nicht mehr in der Lage zwischen Geruchstoffen von Pfirsich und Reinluft (Kontrolle) zu unterscheiden. Mögliche Gründe für diesen Verlust der olfaktorischen Fähigkeiten werden diskutiert. Die genetische Heritabilität der olfaktorischen Fähigkeit war sehr niedrig (0.103). Dies deutet darauf hin, dass die olfaktorischen Reaktionsfähigkeiten nichtgenetisch beeinflusst ist. Die Ergebnisse zeigen deutlich, dass in zukünftigen Studien ausschließlich Feldpopulationen zu verwenden sind.The oriental fruit moth, Grapholita (Cydia) molesta Busk (Lepidoptera: Tortricidae), is one of the most destructive and economically important pests of stone and pome fruits worldwide. Peach is considered to be the primary host of this moth, but also apple and pear have become increasingly important as late season hosts in the past decades. The attraction of G. molesta mated females to their host plant is primarily guided by olfactory cues. Rearing insects in laboratory colonies for many years may cause changes in their olfactory response. Thus, effects of long-term rearing on the olfactory response in G. molesta female moths to plant derived odors were tested. Two G. molesta strains, one that had been maintained in the laboratory for over 70-80 (‘2007 strain) generations and the other for no more than 15 (‘2009 strain) generations, were compared. Also a full-sib/ half-sib approach was used to evaluate if the olfactory responses in this species have a genetic component. Results showed that long-term laboratory rearing affects the olfactory responses of the moths. Females from the ‘2007 strain were unable to discriminate between peach-derived odors vs. clean air (control). In contrast, females from the ‘2009 strain were attracted to the plant-derived odors. Possible reasons for the decreased olfactory abilities caused by rearing are discussed. Estimates of the narrow-sense heritability of olfactory responses were very low (0.103), indicating that non-genetic factors determine such responses.
