Estudo da função da proteína Tau nos mecanismos da neuropatia periférica

Dissertação de mestrado em Ciências da SaúdeNeuropathic pain is defined as the pain evoked by a lesion or disease of the somatosensory system and evidence from several human and animal studies demonstrate that this condition is commonly associated with cognitive and mood deficits. However, the underlying cellular mechanisms of the pain-triggered behavioral deficits are still poorly understood. Recently, several studies suggested that the cytoskeletal protein Tau is a regulator of neuronal plasticity and malfunction in a broader spectrum of brain disorders. For instance, Tau accumulation and hyperphosphorylation represent key events in the neuronal damage found in several neuropathological conditions such as Alzheimer’s disease (AD), epilepsy and chronic stress with Tau reduction blocking the neuronal and behavioral deficits found in the above pathologies. In light of the above evidence and of a recent study demonstrating the essential role of Tau in pain circuitry, the present Master thesis assesses the potential involvement of Tau and its malfunction in neurostructural alterations and brain damage induced by peripheral neuropathy. Using the Spared Nerve Injury (SNI) model of peripheral neuropathy, it is demonstrated that chronic SNI triggers increased transcription of Tau gene as well as Tau accumulation and hyperphosphorylation followed by hippocampal neuronal atrophy and memory impairment. Importantly, analgesic treatment with gabapentin, considerably improves the cognitive performance of SNI animals. Furthermore, in contrast to their wild type littermates, mice lacking Tau (Tau knockout; Tau-KO) that were submitted to SNI exhibit no hippocampal atrophy and related cognitive deficits suggesting that Tau ablation block the establishment of neurostructural and behavioral deficits in hippocampus triggered by SNI model of peripheral neuropathy. Altogether, the findings of this Master thesis add to our limited knowledge about the cellular mechanisms through which peripheral neuropathy evokes brain damage and memory impairments providing the first evidence of the essential involvement of Tau and its malfunction in SNI-driven morphofunctional hippocampal deficits.A dor neuropática é definida como a dor provocada por uma lesão ou doença do sistema somatosensorial. Estudos em humanos e com animals demonstram que esta condição é communmente associada com défices cognitivos e alterações no comportamento emocional. No entanto, os mecanismos celulares subjacentes aos défices comportamentais causados pela dor são ainda pouco compreendidos. Recentemente, vários estudos sugeriram que a proteína citosquelética Tau é um regulador da plasticidade e do mau funcionamento neuronal num amplo espectro de doenças cerebrais. Particularmente, a acumulação e hiperfosforilação da Tau representam eventos-chave nos danos neuronais encontrados em várias condições neuropatológicas tais como a doença de Alzheimer (DA), epilepsia e stress crónico, uma vez que a redução da Tau bloqueia os défices neuronais e comportamentais encontrados nas patologias acima referidas. Considerando as evidência apresentadas assim como um estudo recente que demonstra o papel essencial da Tau nos mecanismos adjacentes aos circuitos da dor, esta tese de Mestrado avalia o potencial envolvimento da Tau e do seu mau funcionamento nas alterações neuroestruturais e no dano cerebral induzido pela neuropatia periférica. Utilizando o modelo de neuropatia periférica Spared Nerve Injury (SNI), é demonstrado que o SNI está associado a um aumento na transcrição do gene da Tau assim como uma acumulação e hiperfosforilação desta proteína seguidos por atrofia neuronal no hipocampo e dano cognitivo. É ainda importante referir que o tratamento de animais SNI com o analgésico gabapentina melhorou significativamente a sua performance cognitiva. Além disso, em contraste com os murganhos wildtype, murganhos com a deleção da Tau (Tau knockout; Tau-KO) que sofreram SNI demonstraram ausência de atrofia no hipocampo assim como dos défices cognitivos associados, sugerindo que a deleção da Tau bloqueia o estabelecimento dos défices neuroestruturais e comportamentais desencadeados pelo SNI no hipocampo. De um modo geral, as evidências apresentadas nesta tese de Mestrado vêm enriquecer o nosso conhecimento limitado acerca dos mecanismos celulares a partir dos quais a neuropatia periférica induz o dano cerebral e a perda de memória, sendo a primeira evidência do envolvimento indispensável da Tau e do seu mau funcionamento nos défices morfofuncionais desencadeados pelo SNI no hipocampo.The work presented in this thesis was performed in the Life and Health Sciences Research Institute (ICVS), Minho University.
