Various types of agents constantly assault DNA. DNA double-strand breaks (DSBs) are the most toxic lesions because their unfaithful repair leads to chromosomal translocations, and cancer. Heterochromatin (HC), which is highly condensed and restricts DNA transactions, makes the DSB repair a challenging process. Previously it was shown that DNA repair in mouse pericentromeric HC is spatially and temporarily uncoupled. In G1, DSBs are positionally stable and recruit NHEJ factors. In S/G2, DSBs are resected and relocate to the periphery of HC where they are retained by RAD51.
