In eukaryotic cells, the degradation of the mRNA poly(A) tail (deadenylation) is a crucial step in the regulation of gene expression. The Ccr4-Not complex is the major deadenylase enzyme involved in the mRNA deadenylation. The complex is composed by two subunits with ribonuclease activity (Caf1 and Ccr4) and at least six non-catalytic subunits. In vertebrate cells, the duplication of the catalytic subunits Caf1 (encoded by CNOT7 and CNOT8) and Ccr4 (encoded by CNOT6 and CNOT6L) lead to the heterogeneity of the complex.
