Développement et évaluation in vitro, ex vivo et in vivo de formulations innovantes d'argiles afin de proposer un nouveau dispositif de décontamination cutanée

The contamination of civilian and military populations through the use of chemical weapons is a current and important risk. In addition, exposure to toxic industrial chemical released into the atmosphere during accidents can also be a source of systemic contamination. Due to their physicochemical properties, some agents will cause contamination through the skin. Decontamination must be carried out as soon as possible to remove the contaminating agent and thus reduce its penetration through the skin. Various products can play this role, including Fuller’s earth, a clay with a high adsorbent capacity when used in powder form. The objective of this work was to develop an innovative formulation based on Fuller’s earth, in order to improve its decontaminating power and broaden its field of use.By setting up an ex vivo diffusion methodology with the Vitropharma® device, the study of the penetration of organophosphorus and mustard gas simulants was possible. Thus the evaluation of the decontamination efficiency of aqueous suspensions of Fuller’s earth made it possible to validate pig skin as a predictive model of human skin for percutaneous penetration studies. The comparison of the decontamination power of different clays in powder and suspension forms, as well as the use of different simulants, revealed a relationship between decontamination, specific area and swelling capacity of the clay. Furthermore, the importance of the stratum corneum as a reservoir compartment for the contaminant was reconfirmed. L’objectif de ce travail a consisté à développer une formulation innovante à base de terre de foulon, afin d’améliorer son pouvoir décontaminant et d’élargir son champ d’utilisation. Par la mise en place d’une méthodologie de diffusion ex vivo avec le dispositif Vitropharma®, l’étude de la pénétration de simulants de composés organophosphorés et de l’ypérite a été possible. L’évaluation de l’efficacité de décontamination de suspensions aqueuses de terre de foulon a ainsi permis de valider la peau de cochon comme modèle prédictif de la peau humaine pour les études de pénétration percutanée. La comparaison du pouvoir décontaminant de différentes argiles sous forme de poudres et de suspensions, ainsi que l’utilisation de différents simulants ont mis en évidence une relation entre décontamination, surface spécifique et capacité de gonflement de l’argile.
