Analysis of a new mitral valve dystrophy animal model : the FlnA-P637Q KI rat : Involvement of immune cells

La dystrophie valvulaire mitrale (DVM) touche 2 à 3% de la population. Néanmoins, aucun traitement pharmacologique n’est disponible actuellement, la seule option thérapeutique est la chirurgie valvulaire. Afin d’étudier la physiopathologie de la DVM, j’ai analysé un nouveau modèle animal : le rat Knock-In (KI) FlnA-P637Q qui exprime la mutation Filamine-A identifiée par notre groupe dans une famille. Grâce à une approche d’imagerie multimodale, j’ai réalisé le phénotypage valvulaire du rat KI FLnA-P637Q, par échocardiographie, microCT et histologie. J’ai ainsi pu démontrer la pertinence de ce modèle, qui développe un DMV de type myxoïde, pour étudier les mécanismes impliqués dans la DVM. L’analyse transcriptomique réalisée à 3 semaines, a mis en évidence l’implication de multiples voies de signalisation dans la pathologie : organisation de la matrice extracellulaire, migration des cellules épithéliales, réponse au stress mécanique et l’implication des cellules immunitaires. Afin de déceler le rôle des cellules immunitaires dans l’initiation et/ou la progression de la pathologie, des approches de cytométrie et qPCR ont été mises en place, conjointement à un phénotypage de la DVM chez des rats âgés de 7 jours. Ces analyses mettent en évidence que le développement de la DVM précède le recrutement des cellules immunitaires.. Ce travail démontre la pertinence du modèle de rat KI FlnA-P637Q dans l’étude de la physiopathologie mitrale et décèle un rôle des cellules myéloïdes dans la pathologie.Mitral valve dystrophy (MVD) touches 2 to 3% of the population. However, there is no pharmacological treatment available, patients are referred to surgery. To study the pathophysiological mechanisms involved in MVD, I analyzed a new animal model: the FlnA-P637Q KI rat which expresses a Filamin- A mutation found by my group in MVD patients. Using multimodal imaging approach based on echocardiography, microCT and histology, I phenotyped the model. I showed the pertinence of the FlnA rat which develops myxomatous dystrophy, to study the mechanisms implied in MVD. 3-week-old MV underwent transcriptomic analysis: extracellular matrix organization, epithelial cell migration, response to mechanical stress, and a central contribution of immune cells are highlighted as the main signaling pathways. To decipher immune cell role in the initiation and progression of MVD, cytometry and qPCR approached have been developed. The analysis reveal a role for myeloid cells in the progression rather than the onset of the pathology. This work reveals the pertinence of the new FlnA-P637Q KI rat model to study mitral pathophysiological processes and highlights a role for myeloid cells in MVD
