In this study, by using peritoneal macrophage and J774A.1 mononuclear macrophage in vitro and dextran sulfate sodium (DSS)-induced colitis mice model in vivo, we found that disulfiram with Cu2+ could significantly reverse the symptom of dextran sulfate sodium (DSS)-induced ulcerative colitis, such as weight loss, disease activity index score; shorten the colon length, and reverse the pathological changes of mouse colon. Disulfiram with Cu2+ could not only inhibit the activation of colonic macrophages by blocking NF-KappaB pathway and reducing NLRP3-inflammasome-derived IL-1β secretion and CASP1 activation, but also reduce IL-17 secretion of CD4+ T cells.
