Efecto de diferentes niveles de Prostaglandina (Dinoprost y Cloprostenol) y Glicosaminoglicanos (Ácido hialurónico) en la motilidad, postdescongelación de espermatozoides bovinos

The aim of this study was to evaluate the effect of two levels: prostaglandin and glycosaminoglycans in sperm motility. A total of 90 samples were obtained from three adult male Gyr breed (animals from The Biotechnological Center of Caribe, CBC, SENA, Valledupar, Colombia) divided into 6 treatments and 5 replicates per treatment. All ejaculated semen was evaluated, divided into 6 aliquots and then, frozen by means of a standard protocol using the commercial diluent, AndroMed®. After freezing, sperm motility was evaluated by means of computer-assisted sperm analysis (CASA). The control treatment and the cloprostenol treatment (0.1 mL) showed statistically higher values (P = 0.05) for most of the evaluated seminal parameters: motility (68.71 ± 26.3% and 12.9 ± 60.80 %, respectively); and progressiveness (18.75 ± 17.01 ± 11.3% and 6.9%, respectively). Hyaluronic acid treatment (0.1 uL) showed the best values in the parameters of curvilinear (VCL) and average path velocity (VAP), and the mean amplitude of lateral sperm head (ALH) displacement. For straight-line velocity (VSL), linearity index (LIN), straightness index (STR), wobble (WOB) and beat cross frequency (BCF), there were no statistical differences (p =0.05). In conclusion, the data obtained in the study reflects sperm damage due to cryopreservation of bovine sperm at the CBC.El objetivo del presente estudio fue evaluar el efecto de dos niveles: prostaglandina y glicosaminoglicanos en la motilidad espermática. Un total de 90 muestras fueron obtenidas de 3 machos adultos de la raza Gyr (animales del Centro de Biotecnología del Caribe CBC; SENA, Valledupar) divididas en 6 tratamientos y 5 repeticiones por tratamiento. Todos los eyaculados fueron evaluados y luego divididos en 6 alícuotas y congelados mediante un protocolo estándar empleando el diluyente comercial Andro-Med®. Después de la congelación, se evaluó motilidad espermática (mediante análisis espermático asistido por computadora CASA). El tratamiento control y el tratamiento de 0,1 µL de cloprostenol presentaron valores estadísticamente (P=0,05) superiores para la mayoría de parámetros seminales evaluados: Motilidad (68,71±26,3 % y 60,80±12,9 %, respectivamente); y progresividad (18,75±11,3 % y 17,01±6,9 %, respectivamente). El tratamiento de 0,1 µL ácido hialurónico presentó los mejores valores en los parámetros de velocidad curvilínea (VCL), velocidad media (VAP) y la amplitud media del desplazamiento lateral de la cabeza del espermatozoide (ALH). Para los parámetros velocidad rectilínea (VSL), índice de linealidad (LIN), índice de rectitud (STR), índice de oscilación (WOB), y frecuencia de batido de cabeza (BCF), no existieron diferencias estadísticas (P=0,05). Concluyendo, los datos obtenidos en el estudio reflejan daños espermáticos debido al proceso de criopreservación de espermatozoides bovinos en el CBC
