Standardization of a PCR-SSP for the identification of phytopathogen phythophtora infestans (Oomycete) from DNA samples in fruits inoculated in eggplant (Solanum Melongena)

La infección causada por Phytophtora infestans ocasiona la enfermedad de gota, marchitez, gotera, lancha o tizón tardío que afecta a la mayoría de especies de la familia Solanaceae. En la berenjena (Solanum melongena) aún no se tiene claro cuál es la especie, o las especies del género Phytophtora que afectan a la planta y que es la causante de la podredumbre de los frutos, y, muchas veces, llega a ser confundido con otros géneros de hongos como Fusarium spp y Botrytis spp. En consecuencia, no se puede determinar qué tipo de control se puede utilizar frente al patógeno y que tipo de funguicida emplear. Inoculando hasta 25 frutos de berenjena utilizando el linaje clonal EC-1, se realizaron extracciones de ADN, que luego fueron cuantificadas por el equipo Nanodrop 2000c; para estandarizar una PCR-SPP empleando Cebadores o Primers de Secuencias ITS. Los resultados de estas extracciones y de las PCRs, fueron visualizadas en geles de agarosa al 2% y 3%. Para probar la sensibilidad de la prueba, se realizaron diluciones de hasta 10-3. El linaje Clonal de Phytophtora infestans EC-1, no está reportado como agente causal de la enfermedad de gota, marchitez, gotera, lancha o tizón tardío en la berenjena, se propone como nuevo patógeno para Solaum melongena. Se utilizaron 3 juegos de primers de secuencias ITS, solo dos de ellos amplifican el tamaño real de una ITS y cumple con sus características.
