Desain Imuno-informatika Vaksin Berbasis Sel T Sitotoksik Terhadap Daerah Lestari Protein Spike Glikoprotein SARS-CoV-2

Background Coronavirus Disease 19 (COVID-19) was first confirmed in the Wuhan area, China since December 31, 2019, the spread of this virus is increasing so that on March 11, 2020, the coronavirus is not a stable virus and can mutate and produce new variants, namely alpha , beta, gamma, and delta which are known to be more virulent than other types of variants. Thus it is necessary to treatment in the form of a vaccine to provide a defense in the form of the vaccine to the body to be more effective in preventing the spread of the virus and provide protection from infection development of vaccine candidate peptides with the base epitope using in silico which has an advantage in predicting the antigen to be useful as a candidate candidate vaccine and is able to produce a strong immune response with minimal side effects so that it is safer to use and more effective, efficient, affordable and effective against all mutations of the Covid-19 virus. The purpose of this study was to find an epitope candidate for the SARS-CoV-2 virus S protein which has potential as a peptide vaccine through in silico review. Method Observational researchthat is descriptive in silico on the SARS-CoV-2 virus vaccine candidate using the Windows 10 home single language 64-bit operating system and the software used is MEGA X, IEDB, VaxiJen 2.0, BLASTp. Results and Conclusions 183 obtained sequences were SARS-CoV-2 virus, then there were 10 candidate epitopes that had antigenic properties and had no resemblance to surface proteins or passed the similarity test so that they have the potential to develop peptide vaccines including TTEILPVSM (score: 1.2262, ILDITPCSF ( score: 1.1835), KTSVDCTMY (score: 1.1824), GAEHVNNSY (score: 0.9347), WTAGAAAYY (score: 0.6306), TSNQVAVLY (score: 0.4387), HADQLTPTTW (score: 0.3807), RVDFCGKLLLGY (score: 0.3717), ECSN score: 0.3331) and lastly LTDEMIAQY (score: 0.1043)
