Synthesis and chemical characterization of new compounds to create bismut (III) chloride in thioamide group and investigation of inhibition effects on lipoxygenase (LOX) enzyme

Bizmut metali diğer ağır metallere göre daha düşük toksisite değerine sahiptir. Bu özelliğinden dolayı bizmut metali tıbbi uygulamalarda daha çok tercih edilir duruma gelmiş ve geniş bir kullanım alanına sahip olmuştur. Tiyoamid türevi ligandlar ise, tek başlarına gösterdikleri tıbbi uygulamalarda ki anti-tümör aktivitelerinin dışında metallerle oluşturdukları bileşiklerinin rahim kanseri (HeLa) gibi bazı kanser hücreleri üzerinde oldukça etkin özelliğe sahip olduğu yapılan literatür çalışmaları ile belirlenmiştir. Araştırmalar sonucu özellikle bizmut(III) halojenürlerin tiyon grubu içeren ligandlar ile oluşturduğu bileşiklerinin kanser hücreleri üzerinde oldukça aktif oldukları belirlenmiştir. Bu tez çalışması kapsamında tiyoamid grubu ligandları olan 2-merkapto-1-metilimidazol, 2-merkaptobenzimidazol, 3,4,5,6-tetrahidro-2-pirimidintiyol, 2-merkaptopiridin ve 2-merkaptobenzotiazol ile bizmut(III) klorür metalinin belirli çözücüler içerisindeki reaksiyonu sonucu oluşan yeni bizmut bileşiklerinin sentezi yapılmış ve kimyasal yapıları aydınlatılmıştır. Kimyasal yapı analizinde erime noktası, iletkenlik, elementel analiz, Mid-IR spektroskopisi, FT-Raman spektroskopisi, Termogravimetrik-Diferansiyal Termal analiz (TG-DTA), 1H ve 13C NMR spektroskopisi, UV spektroskopisi ve X-ışını kırınımı yöntemleri kullanılmıştır. Ayrıca sentezlenen yeni bizmut bileşiklerinin lipoksigenaz (LOX) enzimi üzerindeki inhibisyon etkileri incelenmiştir.Bismuth metal has lower toxicity than other heavy metals. Because of this feature, bismuth metal has become more preferred in medical applications and has a wide usage area. Thioamide-derived ligands have been identified by literature studies that show that compounds other than anti-tumor activity in medical applications that they demonstrate alone have a very effective property on certain cancer cells, such as cervical cancer (HeLa), in their metal-forming compounds. Research results have shown that especially bismuth (III) halides formed with ligands containing tion groups are highly active on cancer cells. In this thesis, we have investigated the effect of bismuth (III) complexes with the thioamide group ligands, 2-mercapto-1-methylimidazole, 2-mercaptobenzimidazole, 3,4,5,6-tetrahydro-2-pyrimidinethiol, 2-mercaptopyridine and 2-mercaptobenzothiazole. The reaction of the bismuth chloride metal in certain solvents has resulted in the synthesis of new bismuth compounds and their chemical structures are elucidated. Chemical structure analysis, melting point, conductivity, elemental analysis, Mid-IR spectroscopy, FT-Raman spectroscopy, Thermogravimetric-differential thermal analysis (TG-DTA), 1H and 13C NMR spectroscopy, UV spectroscopy, and X-ray diffraction methods were used. In addition, bismuth compounds newly synthesized by lipoxygenase (LOX) enzymes on the inhibition effects were investigated
