This paper describes attempts to identify parameters that are critical for succesful gene transfer into the germ line of mice by microinjection of foreign DNA into fertilized eggs. in the first stage of the study, 1470 zygotes were obtained and 1269 (86%) were microinjected with a human growth hormone gene construct. Of these zygotes, 769 (60%) proved to be alive after the injection, and 420 of them were transfered into the pseudopregnant recipients (5496). As a result, 70 offsprings were obtained, and out of these 3 (4%) proved to be transgenic.
