A batch procedure for purifying native and for separating single-stranded fractions from alkali-denatured DNA of Bacillus subtilis, employing MAK column chromatography, was described. Hydroxyapatite chromatography was utilized in the physical characterization of the single-stranded fractions. Biological characterization of the single-stranded fractions, either using them alone or reannealed in various combinations in transformation assays, demonstrated an enrichment for particular genetic traits. A difference detected in the elution profiles and transforming activity of the single-stranded fractions from an auxotroph (Ade6) and a prototroph of B. subtilis 168 implied an apparent physical difference in the constituent DNA of the strains.
