Dans ce travail de thèse, nous avons étudié le mode d’action in vitro de différentes classes de perturbateurs endocriniens (PE) sur l’activité du récepteur aux acides gras couplés aux protéines G : GPR40. Nous nous sommes intéressés aux bisphénols (BPA), aux polluants organiques persistants (le pesticide organochloré DDT et son métabolite DDE) ainsi qu'aux phtalates (DEHP et son métabolite MEHP). Nous avons caractérisé le récepteur GPR40 comme une cible des PE. Nous avons mis en évidence que chaque composé a un mode d'action différent des autres : le BPA module négativement la réponse au GW9508 (ligand synthétique) mais n'a aucun effet sur celle de l’acide linoléique (ligand endogène) ; le DEHP potentialise la réponse au GW9508 et diminue c...
