Durch die Kristallisation des Enzyms im Komplex mit dem Inhibitor 5-Flourlävulinsäure konnten die Struktur des mit Substrat beladenen Enzyms zum Zeitpunkt des Starts der Katalyse, sowie die ersten Schritte im Reaktionsverlauf modelliert werden. Ein Reaktionsmechanismus konnte formuliert werden, der die Aktivierung des Substrats in der A-Stelle in Form eines Enamins, die Bildung der C-C-Bindung als erstem Reaktionsschritt und die Beteiligung eines monovalenten Kations beinhaltet.
