The Radical SAM enzyme oxygen-independent coproporphyrinogen III oxidase (CPO) catalyzes the oxidative decarboxylation of coproporphyrinogen III to protoporphyrinogen IX during heme and chlorophyll biosynthesis. This work included further characterization of Escherichia coli HemN and investigation of two proposed alternative oxygen-independent CPOs, designated HemZ and HemZ2. Structure-based mutagenesis of E. coli HemN was performed to investigate the functional role of the second S-adenosyl-L-methionine (SAM) molecule bound in the HemN crystal structure (SAM2) and the proposed HemN substrate binding site.
