Formulation of oil-in-water emulsions with combinations of hypromellose and characterization of their kinetics with respect to creaming, coalescence, and flocculation, using a novel laser-photometer

In dieser Arbeit werden Emulsionen untersucht, die mit Hypromellose (HPMC) stabilisiert sind. Ziel ist es, durch Kombination von kurz- und längerkettigen HPMC, verschiedenen Substitutionstypen und Variation der Herstellungsmethode, feindisperse und lagerstabile Emulsionen zu erzeugen. Betrachtet werden Emulsionen mit einem Polymer und mit Polymerkombinationen. Verglichen werden zudem die gemeinsame und sequentielle Verarbeitung der Polymere. Es werden die Tropfengrößenverteilung (TGV), Viskosität und Aufrahmstabilität der Zubereitungen nach Herstellung und bei Lagerung untersucht. Für die Beobachtung der Aufrahmstabilität wird ein Laser-Photometer als neue in-situ Methode verwendet. Dieses Laser-Photometer wird hier erstmalig systematisch auf seine Eignung für die Stabilitätsanalytik von Emulsionen geprüft. Es wird gezeigt, dass mit dem Laser-Photometer schnelle und gut reproduzierbare Aussagen zur Aufrahm- und Koaleszenzstabilität ohne vorherige Probenpräparation von Emulsionen mit einem Innenphasenanteil von bis zu 30 % möglich sind. Die TGV, Viskosität und Aufrahmstabilität der Emulsionen mit Polymerkombinationen wird stark von den Substitutionstypen der HPMC beeinflusst. Die Kombination von Polymeren mit unterschiedlichen Substitutionstypen führt zu reversibler Flockung, induziert durch Verbrückung der Tropfen. Bei gleichem Substitutionstyp tritt hingegen keine Flockung auf, vielmehr entsteht ein gel-ähnliches Netzwerk in der Außenphase. Dies führt bei Verwendung der längerkettigen HPMC als Kombinationspartner zu hoher Aufrahmstabilität. Eine sequentielle Verarbeitung der Polymere des gleichen Substitutionstyps bei geringem Energieeintrag im zweiten Herstellungsschritt führt zudem zu deutlich engeren TGV als bei gemeinsamer Verarbeitung oder Verwendung von einem Polymer allein.In this work, emulsions stabilized with hypromellose (HPMC) are investigated. One goal is formulation of fine disperse and storage stable emulsions by combination of short- and medium-chainlength HPMC. Emulsions with a single polymer and polymer combinations as emulsifiers are considered as well as combined and sequential processing of the polymers. Droplet size distribution (DSD), viscosity, and creaming stability of the formulations are studied after preparation and during storage. For observation of creaming stability, a laser photometer is used as a new in-situ method. The applicability of this laser photometer for stability assessment of emulsions is studied systematically for the first time in the present work. It is shown that the laser photometer provides fast and good reproducible results with respect to creaming and coalescence stability without prior sample preparation of emulsions with an oil volume fraction up to 30%. DSD, viscosity, and creaming stability of emulsions with polymer combinations are strongly influenced by the substitution types of the HPMC used. Combination of polymers with different substitution types lead to reversible flocculation induced by bridging. Polymer combinations with different substitution types build a gel-like network in the continuous phase and show no flocculation. The use of medium-chainlength polymers as combination partner in addition leads to high creaming stability. Sequential processing of polymers of the same substitution type at low energy application in the second processing step leads to remarkable narrow DSD compared to combined polymer processing or use of a single polymer
