Entwicklung eines HTS-fähigen Zellassays basierend auf der proteolytischen Spaltung oder Inhibition des humanen La Proteins

Despite effective vaccines, Hepatitis B still causes over one million deaths worldwide each year. The current drug treatments have adverse effects in patients and are compromised by viral resistance. New methods of treatment avoiding the resistance development are urgently needed. The La protein is a host protein required for the stabilization of Hepatitis B Viral (HBV) RNA. The main objective of this thesis was to develop a platform for efficient screening of chemical libraries with cell based assays targeting the proteolytic cleavage or inhibition of the La protein as read-out using fluorescence microscopy. The concept of the platform involves the in situ solid phase combinatorial chemical synthesis of compounds on a planar array of resin patches polymerised on a hydrophobic polypropylene surface (foil), followed by direct cultivation of the reporter cell line on the patch array with simultaneous release of the compounds from a suitable linker (Frank-Linker). Such a patch screening platform was developed during the course of this thesis. The compatibility of the PP-patch surface displaying diverse chemical properties with the cultivation of different cell lines commonly used for cell based assays was successfully tested. Cellular phenotypes such as lamellipodia induction in 3Y1 cells and the inhibition of cytokinesis in PtK2 cells by in situ synthesised Tat-DT2 peptides were identical to experiments on conventional glass surfaces. This demonstrated the full functionality of the planned screening platform. Concerning the La-targeting cell based assay, initial attempts to measure FRET changes upon cleavage of an ECFP-La-EYFP construct were not sensitive enough. Alternatively, the intracellular translocation of a proteolytic fragment of an N-terminal GFP-NES-La from the nucleus to the cytoplasm proved a much more robust readout. This is now ready for screening on the developed platform using new automated microscopy and image analysis instrumentation.Trotz effektiver Impfstoffe führen Hepatitis B Infektionen noch jedes Jahr zu über einer Million Toten. Die vorhandenen Medikamente zur Behandlung haben erhebliche Nebenwirkungen und führen zu Resistenzbildung. Neue Medikamente, die eine Resistenzbildung vermeiden, werden dringend gebraucht. Das La Protein ist ein Wirtsprotein das die virale RNA des Hepatitis B Virus stabilisiert. Ziel der Arbeit war die Entwicklung einer neuen Technologieplattform für das Screening von Substanzbibliotheken mit zellbasierten Tests, die eine proteolytische Spaltung oder Inhibition des La Proteins durch Fluoreszenzmikroskopie sichtbar machen. Das Konzept der Technologie sieht eine kombinatorische Festphasensynthese von Substanzen auf einem planaren Raster von kleinen Polymerflächen (patches) auf einer hydrophoben Polypropylenfolie vor, auf denen die Testzellen unter gleichzeitiger Ablösung der Substanzen von einem geeigneten Linker (Frank-Linker) direkt kultiviert und nach Fluoreszenzfärbung beobachtet werden können. Die Kompatibilität der unterschiedlich chemisch veränderten Polymeroberflächen mit einer Vielzahl an Zelllinien wurde erfolgreich getestet. Zelluläre Phänotypen wie die Induktion von Lamellipodien in 3Y1 Zellen oder die Inhibition der Cytokinese in PtK2 Zellen durch in situ synthetisierte Tat-DT2 Peptide zeigten sich identisch zu Experimenten auf konventionellen Glasoberflächen. Dies belegt die volle Funktionsfähigkeit der entwickelten Screening-Plattform. Ansätze für einen La-gerichteten Zelltest über die Bestimmung der spaltungsabhängigen Änderungen des FRET-Signals eines ECFP-La-EYFP Konstruktes waren wegen zu geringer Signalstärke nicht erfolgreich. Alternativ ergab die Beobachtung der intrazellulären Translokation eines proteolytischen N-terminal Fragmentes eines GFP-NES-La aus dem Nukleus ins Cytoplasma einen sehr viel robusteren Messparameter. Neue automatisierte Fluoreszenzmikroskope und Bildanalyse stehen für die entwickelte Screening-Plattform zur Verfügung
