Die Rolle von Cortactin in Arp2/3-abhängigen Prozessen und Aktinnukleation

Actin polymerization is a fundamental cellular process driving functions as diverse as cell migration, endocytosis and adhesion. To fulfill its various tasks in the cell, the assembly and disassembly of actin filaments has to be tightly regulated. Cortactin has been described as Arp2/3 activator stabilizing branches in dendritic actin filament networks. The characterization of cortactin KO cells has challenged this view, as lamellipodia formation e.g. was normal in the absence of cortactin. In this thesis, the potential functions of cortactin in migration, podosome formation and on Arp2/3 activation were explored. The observed migration defect in cortactin-deficient cells could be restored by overexpression of constitutively active Rho-GTPases, whose activities were decreased in cortactin KO cells. Cortactin was also found to be essential in InlB-mediated uptake of Listeria, the mechanism of which remains to be determined. Moreover, macrophages from cortactin KO mice were able to form podosomes. Furthermore, the ability of cortactin to directly activate the Arp2/3 complex was tested in vivo. For this purpose, a microtubule-based actin nucleation assay was developed, allowing targeting of actin nucleators or nucleation promoting factors to microtubules. A microtubule-targeting domain fused to the N-WASP-VVCA induced actin polymerization on microtubules in an Arp2/3-dependent fashion. However, cortactin did not drive actin filament assembly on microtubules nor induce accumulation of Arp2/3-complex, consistent with accumulating evidence questioning a direct role for cortactin in Arp2/3 activation. However, cortactin was recruited downstream of Arp2/3-dependent actin assembly activated e.g. by N-WASP. By using the MBD-assay, it was possible to recapitulate all known actin nucleation mechanisms known to date, including those mediated by Spir and formin. It could further be demonstrated that the A-domain of N-WASP is not essential but helpful for Arp2/3 activation.Aktinpolymerisation ist ein fundamentaler, zellulärer Prozess, der essentiell für Migration, Endozytose und Adhäsion ist. Um die unterschiedlichen Aufgaben in der Zelle zu erfüllen, müssen Aufbau und Abbau von Aktinfilamenten stark reguliert sein. Cortactin wurde bislang als Arp2/3-aktivierendes und verzweigungsstabilisierendes Protein beschrieben. Die Charakterisierung von Cortactin KO-Zellen ergab, dass Arp2/3-abhängige Prozesse wie die Lamellipodienbildung in Abwesenheit von Cortactin nicht beeinträchtigt sind. In dieser Arbeit wurden die Auswirkungen von Cortactin auf die Migration, Podosomenbildung und Arp2/3-Aktivierung untersucht. Der beobachtete Migrationsdefekt in Cortactin-defizienten Zellen konnte durch Überexpression von konstitutiv aktiven Rho-GTPasen rekonstituiert werden, deren Aktivität in Cortactin KO-Zellen verringert war. Cortactin wurde außerdem als essentielle Komponente in der InlB-vermittelten Aufnahme von Listerien ermittelt. Makrophagen aus Cortactin KO-Mäusen konnten Podosomen bilden. Weiterhin wurde getestet, ob Cortactin den Arp2/3-Komplex in vivo aktivieren kann. Dafür wurde ein mikrotubulibasierter Aktinnukleationssassay entwickelt, bei dem Aktinnukleatoren oder nukleationsfördernde Faktoren an Mikrotubuli rekrutiert werden. Die Rekrutierung der N-WASP-VVCA-Domäne an Mikrotubuli zeigte, dass Arp2/3-abhängige Aktinnukleation ektopisch induziert werden kann. In Einklang mit anderen Daten, die eine direkte Rolle von Cortactin auf die Aktivierung von Arp2/3 in Frage stellen, war Cortactin nicht in der Lage, Aktinpolymerisation oder Arp2/3-Rekrutierung an Mikrotubuli auszulösen. Dennoch wurde Cortactin an Aktinfilamente rekrutiert, die vom Arp2/3-Komplex nukleiert wurden. Alle zurzeit bekannten Aktinnukleationsmechanismen, wie Spir- und Formin-vermittelte Nukleation, konnten in vivo im MBD-Assay rekapituliert werden. Weiterhin wurde gezeigt, dass die A-Domäne in N-WASP zwar hilfreich, nicht aber essentiell für die Arp2/3-Aktivierung ist
