Synthesis of trichloromethyl-substituted δ-valerolactones as precursors towards a total synthesis of the natural product sigillin and identification of chlorinated lactones from collembola

In Cuticularextrakten von Ceratophysella sigillata, einer winteraktiven Collembolenart (Springschwänze, Überklasse Hexapoda) war bereits vor Beginn der vorliegenden Arbeit ein pentachloriertes, bicyclisches Lacton nachgewiesen worden, welches Sigillin genannt worden ist, und erste Schritte zur Totalsynthese der postulierten Struktur waren unternommen worden. In der vorliegenden Arbeit konnten durch GC/MS-Untersuchungen Strukturvorschläge für sieben weitere strukturanaloge Verbindungen (Sigillin B – H) und ein Vorschlag zur Biosynthese aufgestellt werden. Es wurden Versuche zur Isolierung eines möglichen Sigillin produzierenden Endosymbionten unternommen. Die zum Lactonring des Sigillins strukturanaloge Verbindung 4-Hydroxy-3-methyl-6-(trichlormethyl)tetrahydro-2H-pyran-2-on wurde über drei verschiedene Syntheserouten dargestellt: Cyclisierung der β,δ-Dihydroxysäure, Michael-Addition von Dimethylphenylsilyllithiumcuprat an das korrespondierende α,β-ungesättigte Lacton mit anschließender Tamao-Fleming-Oxidation und Samariumdiiodid-vermittelte intramolekulare Reformatsky-Reaktion von 1,1,1-Trichlor-4-oxobutan-2-yl 2-brompropanoat. Zum ersten Mal konnte auch eine Ytterbiumdiiodid-vermittelte Reformatsky-Reaktion durchgeführt werden. Zum späteren Aufbau des zweiten Rings sollten terminale Alkenylgruppen in α- und β-Position des Hydroxylactons eingeführt werden. Das β-homoallylsubstituierte Hydroxylacton wurde erneut per Reformatsky-Reaktion hergestellt, der Aufbau des α-vinylsubstituierten Hydroxylactons durch vinyloge Reformatsky-Reaktion war jedoch nicht erfolgreich (Eliminierung). Es konnte aber durch Lactonisierung des α-vinylierten Esters dargestellt werden. Der Versuch einer Einführung des zweiten Alkenylsubstituenten in β-Position führte beim α-Vinyl-β-ketolacton zu einer Grignard-Reduktion, und beim offenkettigen Ester zu einer Retro-Aldol-Reaktion. Abschließend wurden verschiedene mögliche Lösungsansätze zu einer späteren Synthese des Sigillins diskutiert.Prior to this thesis, a pentachlorinated, bicyclic lactone had been identified in cuticular extracts of Ceratophysella sigillata, a winter-active collembolan (springtail, subphylum hexapoda), and first steps towards a total synthesis of the proposed structure, which was termed sigillin, had been made. In this thesis, structures were proposed for seven analogous compounds (sigillin B – H). An isolation of a possible sigillin-producing endosymbiont was attempted. The lactone 4-hydroxy-3-methyl-6-(trichloromethyl)tetrahydro-2H-pyran-2-one, which resembles the lactone moiety in the proposed structure of sigillin, was synthesized via three different routes: a direct cyclisation of the corresponding β,δ-dihydroxy ester, a Michael addition of dimethylphenylsilyllithium cuprate to the corresponding α,β-unsaturated lactone with subsequent oxidative Tamao-Fleming cleavage, and a samarium diioidide mediated intramolecular Reformatsky reaction of 1,1,1-trichloro-4-oxobutan-2-yl 2-bromopropanoate. In optimization studies a previously unknown ytterbium diiodide mediated Reformatsky reaction was performed. The introduction of alkenyl sidechains in α- and β-position of the hydroxylactone is crucial for the subsequent formation of the second ring of sigillin via a ring-closing metathesis reaction. A synthesis of the β-homoallyl hydroxylactone was achieved via Reformatsky reaction, but a synthesis of the α-vinyl hydroxylactone via a vinylogous Reformatsky reaction was not successful due to elimination. However, it was successfully synthesized by lactonisation of the preceding α-vinyl ester, which was obtained from ethyl α-methyl-α-vinylacetoacetate. Attempts towards an introduction of the second alkenyl substituent in β-position via Grignard reaction led to a Grignard reduction in the α-vinyl ketolactone and to a retro-aldol reaction in the corresponding open-chain ester. Finally, possible solutions towards a later total synthesis of sigillin were suggested
