Prenylumlagerungen an Indolalkaloiden aus dem marinen Moostierchen Flustra foliacea: Synthese von Flustramin A und Untersuchungen zur Synthese von ent-Flustramin C und Debromflustramin E

Flustra foliacea is a marine bryozoan known as a rich source for indole alkaloids. The chemical synthesis of Flustra alkaloids and biological evaluation of the synthetic natural products and analogues was explored in this work. Deformylflustrabromine was subjected to Lindel’s oxidative 2-tert-prenyl rearrangement. On reaction with NBS, cyclization and sigmatropic [1,5] rearrangement of the 2-tert-prenyl group was induced to result in rac-flustramine C. No brominated side products were detected. The individual enantiomers of rac-flustramine C were separated and the optical rotation was determined to be [alpha]D 21.6 = +153 ° (c = 7.9 mg/mL, CHCl3) and [alpha]D 21.8 = –155 ° (c = 8.1 mg/mL, CHCl3), respectively. Reaction of deformylflustrabromine with NBS, NCS, and NIS furnished rac-flustramine C in every case. The hypervalent iodine reagent PIFA formed rac-flustramine in low yield (5%). Rac-flustramine C was reduced to rac-dihydroflustramine C. Lindel’s rearrangement also furnished rac-debromoflustramine C, and reduction led to rac-debromodihydroflustramine C for the first time. The scope of Lindel’s rearrangement reaction was evaluated. The bromine substituent on benzene section is not required. The methyl group at indole N-1 or an aliphatic amine do not disturb the cyclization/prenyl shift sequence whereas presence of the proton at the side-chain amine is necessary. However, if the side chain beared an amide or a tertiary amine moiety, no formation of pyrrolo[2,3-b]indoles was observed. Na-Prenyl-deformylflustrabromine on reaction with NBS formed the amidinium salt, which was reduced to rac-flustramine A in a yield of 28% over the last two steps. Na-Prenyl-deformylflustrabromine showed cytotoxicity with a geometric mean IC50 value of 5.14 µM against 42 human cancer cell lines. Na-prenyl-deformylflustrabromine and its debromo analogue showed IC50 values of 5.9, 7.7 µM, and 22.6, 7.7 µM against the gram positive bacteria Micrococcus luteus and Mycobacterium phlei, respectively. None of the 30 tested compounds were antifungal. 2-Prenyl-phthaloyltryptamine inhibited the bacterial biofilm formation at concentrations of 1, 5, and 25 µM, respectively.Flustra foliacea ist ein marines Moostierchen und bekannt als ergiebige Quelle von Indol-Alkaloiden. Die Synthese dieser Flustra Alkaloide, sowie die Untersuchung der biologischen Aktivitäten der synthetisierten Naturstoffe und deren Analoga sind in der vorliegenden Arbeit dargestellt. Durch Lindels oxidative, NBS induzierte sigmatrope [1,5] Umlagerung der 2-tert-prenyl Gruppe bei gleichzeitiger Zyklisierung konnte rac-Flustramin C ausgehend von Deformylflustabromin synthetisiert werden. Dabei kam es nicht zur Bildung von bromierten Nebenprodukten. Die Trennung der Enantiomere ermöglichte die Bestimmung der optischen Aktivität der einzelnen Verbindungen mit Drehwerten von +153 ° bzw. –155 °. Desweiteren konnte diese Reaktion auch mit NCS, NIS und PIFA erfolgreich durchgeführt werden, wobei die Ausbeute mit PIFA lediglich 5% betrug. Rac-Flustramin C konnte zu rac-Dihydroflustramin C reduziert werden. Ebenso gelang Lindels Umlagerung mit rac-Debromflustramin C, gefolgt von der Reduktion zu rac-Debromdihydroflustramin C. Weiterhin wurden die Substratbandbreite für Lindels Umlagerung untersucht. Brom am Benzolteil des Indols ist nicht notwendig. Methylierung sowohl des Indol-stickstoffs, als auch des Amins der Seitenkette sind in der Reaktion toleriert. Allerdings ist ein Proton am Amin notwendig. Amide und tertiäre Amine in der Seitenkette können nicht zu Pyrrolo[2,3-b]-Indolen zyklisiert werden. Na-Prenyl-deformylflustrabromin bildet bei Reaktion mit NBS ein Aminidium-Salz, welches zu rac-Flustramin A reduziert werden kann (28% Ausbeute über die letzten zwei Stufen). Na-Prenyl-deformylflustrabromin zeigt Zytotoxizität mit einem mittleren IC50 von 5.14 µM gegenüber 42 humanen Krebszelllinien. Na-Prenyl-deformylflustrabromin und das nicht bromierte Analogon zeigten weiterhin Aktivität gegenüber den grampositiven Micrococcus luteus (IC50 = 5.9 bzw. 7.7 µM) und Mycobacterium phlei (IC50 = 22.6 bzw. 7.7 µM). Keine der 30 getesteten Substanzen zeigte antifungale Aktivität. 2-Prenyl-phthaloyltryptamin inhibierte die Bildung von bakteriellen Biofilmen bei Konzentrationen von 1, 5 und 25 µM
