Heterogeneity in transgene expression is frequently observed upon genetic modification of cells in basic research and biotechnology. The variable transgene expression is considered to be a result of the crosstalk of the incoming promoter cassette with cis-acting elements associated with the chromosomal site of transgene integration (position effect). Targeted integration of the transgene into the open chromatin reduces variability due to chromosomal position effects and also favors the more predictable transgene expression.
