Natural products with oxidized chroman partial structure: total synthesis of the flavonol morin from the mulberry tree Maclura tinctoria and studies towards the synthesis of Dankastatin C from the fungus Gymnascella dankaliensis

Im Zentrum der Dissertation stehen zwei Naturstoffe mit oxidierter Chroman-Teilstruktur, das Flavonol Morin aus dem Färbermaulbeerbaum Maclura tinctoria und der Tyrosin-Abkömmling Dankastatin C aus dem marinen Pilz Gymnascella dankaliensis. Morin wird noch heute aus dem tropischen Färbermaulbeerbaum isoliert und kann verunreinigt sein. Um eine Abhängigkeit von Tropenholz zu verringern und ein reines Morin verfügbar zu machen, war ein Ziel dieser Arbeit, eine Laborsynthese zu entwickeln, die sich auch für den Multigramm-Maßstab eignete und möglicherweise technisch umsetzen ließe. Die Syntheseroute beginnt mit kommerziell erhältlichem Phloroglucin, welches in konzentrierter Schwefelsäure und Methanol zweifach methyliert wurde. Eine umfangreiche zur Monoacetylierung von 3,5 Dimethoxyphenol ergab, dass die Kombination aus Essigsäure und P2O5 in Anwesenheit von Kieselgel sich als optimal erwies. Das Rohprodukt wurde mit 2,4 Dimethoxybenzaldehyd zum Chalkon umgesetzt. Bei der Cyclisierung vom Chalkon wurde neben Tetramethylmorin auch das tetramethylierte Auron gebildet, das als sauberes Nebenprodukt abfiltrierbar war. Die Demethylierung von Tetramethylmorin lieferte Morin nach Extraktion mittels HEMWat Lösungsmittelsystem. Die erstmalige Synthese vom marinen Naturstoff Dankastatin C wäre wichtig, da dieser mit einer mittleren effektiven Dosis (ED50) von 57 ng/mL gegen die Zelllinie P388 wirkt und für weiterführende medizinische Tests in ausreichender Menge zur Verfügung gestellt würde. Die Synthese der Seitenkette ist bereits bekannt, wobei hier eine verbesserte konvergente Synthese über fünf lineare Stufen und einer Gesamtausbeute von 47% ausgehend von n Octansäurechlorid entwickelt wurde. Durch Verwendung eines komplexeren Phosphonats, welches die Methylverzweigung an der Doppelbindung direkt einführt, konnte die Syntheseroute verkürzt werden. Dadurch konnten die bisher verwendeten zweifachen WITTIG- bzw HORNER-WADSWORTH-EMMONS (HWE) Reaktionen umgangen werden. Der Bicyclus von Dankastatin C wurde über eine intramolekulare MICHAEL-Reaktion eines α,β ungesättigten Ketons versucht, welches aus L-Tyrosin synthetisiert wurde. Eine Cyclisierung des Ketons oder eines strukturähnlichen Derivats wurde jedoch nicht erreicht. Ein weiterer Versuch, den Bicyclus von Dankastatin C zu erreichen, lief über ein Cumarin, das durch Chlorieren eines Benzaldehyds und anschließender Umsetzung mit N-Acetylglycin erhalten wurde.The focus of the dissertation is on two natural substances with oxidized chromane partial structure, the flavonol morin from the dyer's mulberry tree Maclura tinctoria and the tyrosine derivative Dankastatin C from the marine fungus Gymnascella dankaliensis. Morin is still isolated from the tropical mulberry tree and may be contaminated during its extraction. In order to reduce consumtion on tropical wood and to make pure morin available, the aim of this work was to develop a laboratory synthesis that was also suitable for the multigram scale and could possibly be technically implemented. The synthesis route begins with commercially available phloroglucinol, which has been methylated twice under reflux in concentrated sulfuric acid and methanol. The monoacetylation of 3,5 dimethoxyphenol was investigated in an extensive study. The combination of acetic acid and P2O5 in the presence of silica gel proved to be optimal. The crude product was converted to the chalcone with 2,4-dimethoxybenzaldehyde. In the cyclization of the chalcone, in addition to tetramethylmorin, the tetramethylated auron was also formed, which could be filtered off as a clean by-product. The demethylation of tetramethylmorin provided morin after extraction using the HEMWat solvent system. The first time synthesis of the marine natural product Dankastatin C would be important, since it acts with a median effective dose (ED50) of 57 ng/mL against the cell line P388 and would be made available in sufficient quantities for further medical tests. The synthesis of the side chain is already known. An improved convergent synthesis was developed over five linear steps and a total yield of 47% starting from n-octanoic acid chloride. By using a more complex phosphonate, which directly introduces the methyl branch on the double bond, the synthesis route could be shortened. As a result, the double WITTIG or HORNER-WADSWORTH-EMMONS (HWE) reactions used so far could be avoided. The yield was further increased by isomerization of the by-product of the HWE reaction. The bicyclus of dankastatin C was attempted through an intramolecular MICHAEL reaction of an α,β-unsaturated ketone synthesized from L-tyrosine. However, cyclization of the ketone or a structure-like derivative has not been achieved.
