Intrabodies - Selection from human naïve antibody repertoires

Intrazelluläre Antikörper, Intrabodies, bieten mit Ihrer spezifischen Bindung von Antigenen die Möglichkeit Protein-Interaktionen und zelluläre Prozesse in lebenden Zellen auf dem post-translationalen Level zu untersuchen. Die Anwendung von Intrabodies wird durch ineffiziente Faltung und Stabilität im reduzierenden Milieu des Zytoplasmas limitiert. Ziel der Arbeit war die Etablierung eines Screenings für Antikörperkandidaten auf optimale Intrabody-Eigenschaften. Als Grundlage dienten scFv-Fragmente, die aus der humanen Antikörperbibliothek HAL7/ 8 gegen unterschiedliche intrazelluläre Antigene isoliert wurden. Beim ELISA unter reduzierenden Bedingungen (R-ELISA) wurden die reduzierenden Bedingungen in dem Screening während der Antigenbindung mit DTE eingestellt. Da Intrabodies nicht nur unter reduzierenden Bedingungen ihr Antigen erkennen, sondern auch im Zytoplasma unter diesen gebildet und gefaltet werden müssen, wurden im nächsten Schritt die scFv-Fragmente in E. coli intrazellular produziert und anschließend im ELISA-System getestet. Als Alternative zum Screening vorhandener scFv-Klone wurde bereits bei der Selektion neuer Klone angesetzt. Somit sollten reduzierende Bedingungen beim Panning zur Identifizierung geeigneter Intrabody-Kandidaten führen. Dafür wurden zwei unterschiedliche Panningstrategien unter reduzierenden Bedingungen (R-Ranning und dR-Panning) mit dem Standardpanning unter nicht reduzierenden Bedingungen (NR-Panning) verglichen. Beide Strategien für ein Panning unter reduzierenden Bedingungen erhöhten die Rate an funktionellen Intrabodies nicht signifikant. Desweiteren wurde sowohl die Produktion von Intrabodies in humanen Zellen, sowie die Antikörper-Antigen-Bindung analysiert. Es wurde ein SPLIT-Luc-Assay für Intrabodies entwickelt. Der Nachweis der Antigenbindung mittels SPLIT-Luc-Assay in lebenden humanen Zellen funktioniert für das getestete Antigen. Beim Vergleich aller Methoden fällt der R-ELISA mit vielen falsch positiven Klone auf. Die Intrabody-Produktion in E. coli mit anschließendem ELISA ist die zuverlässigste Variante möglichst viele scFv-Klone zu testen. Der Vergleich der Produktion von Intrabodies in E. coli und humanen Zellen weist identische positive Klone auf und dieses Ergebnis lässt sich im SPLIT-Luc-Assay bestätigen.Intrabodies are intracellular antibodies with promising applications like studying or blocking protein interactions and cellular processes in living cells. Targeting proteins on a post-translational level and the specific binding of antigens are the greatest advantages of intrabodies in comparison with classical knockout strategies. The use of intrabodies may be limited by inefficient folding and stability in the reducing environment of the cytoplasm. The aim of this thesis was a screening method for functional intrabodies. Therefore scFv fragments against different intracellular antigens were used, which are isolated from the human naïve antibody library HAL7/ 8. First, an ELISA under reducing conditions (R-ELISA) was tested as screening method. To mimic the reducing conditions of the cytoplasm DTE was added to the antigen binding step of an ELISA based screening. But intrabodies should not only recognize their under reducing conditions. They also have to be expressed and correctly folded in the cytoplasm. The next step was the intracellular production of different scFv fragments in E. coli. The antigen binding of this intrabodies was checked by titration ELISA. In parallel with screening of existing scFvs, the direct selection of intrabodies by panning was analyzed. Reducing conditions during panning should lead to the identification of suitable intrabody candidates. Two different panning strategies for the antigen PPP1R12B under reducing conditions (R-Panning and dR-Panning) were compared with the standard panning under non-reducing conditions (NR panning). However, panning under reducing conditions does not significantly increase the rate of functional intrabodies. Furthermore the antibody antigen production and binding in living human cells was supposed to be analyzed. Therefore a SPLIT-Luc assay for intrabodies was developed. The detection of antigen binding by means of SPLIT-Luc assay works for anti-ERK2-intrabodies and their antigen ERK2. This is an important step towards the intracellular use of intrabodies. By comparing all methods, the R-ELISA has noticeable many false positive clones. The intrabody production in E. coli with a subsequent ELISA is the most reliable variant to test as many scFv fragments as possible. The comparison of the production of intrabodies in E. coli and human cells shows identical positive clones and this result can also be confirmed in the SPLIT-Luc assay
