Molecular biology applied to genetic studies of Western Atlantic billfish and swordfish: population aspects and forensic techniques for management and conservation.

Os peixes-de-bico (agulhões e espadarte) são distribuídos globalmente. Como grandes predadores, ocupam o ápice da cadeia alimentar, desempenhando um importante papel ecológico no ambiente marinho. Além de serem alvo da pesca esportiva, são grandemente explorados pela pesca comercial. Sua sobreexploração pode resultar em efeitos negativos para a produtividade, estabilidade e estrutura dos ecossistemas marinhos. Dessa forma, dados genéticos são importantes para o planejamento do manejo pesqueiro ao fornecer informações sobre a distribuição, estrutura e diversidade genética das populações, além de ser uma ferramenta altamente precisa para identificar espécies quando caracteres morfológicos diagnósticos não estão disponíveis. O presente trabalho objetivou empreender o primeiro estudo genético populacional de Tetrapturus pfluegeri (agulhão-verde) do Atlântico Ocidental, utilizando marcadores moleculares nucleares e mitocondriais. Ademais, metodologias de genética forense foram aplicadas para se conhecer a composição de peixes-de-bico em comercialização na maior central de abastecimento de produtos agrícolas da América Latina, a CEAGESP (Companhia de Entrepostos e Armazéns Gerais de São Paulo). Com isso, foi verificado que T. pfluegeri é composto por diferentes grupos genéticos, com diferenciação altamente significante (FST = 0,075 - 0,198; p < 0,05) (DEST = 0,092 - 0,176; p < 0,05), decorrente de um possível processo de filopatria reprodutiva. Além disso, das 432 amostras coletadas na CEAGESP 78,01% pertenciam à espécie Xiphias gladius, 20,83% a Istiophorus platypterus e 0,23% a Tetrapturus georgii, fornecendo evidências de que métodos moleculares são altamente eficientes para superar a dificuldade de identificação dos peixes-de-bico e a captura ilegal de espécies sobreexploradas no oceano Atlântico.Billfish and swordfish species are globally distributed. As big predators, they are in the apex of the food chain, with an important ecological role in the marine environment. They are target species in the sport fishing and highly exploited by the commercial fisheries. Its overexploitation can lead to negative effects on productivity, stability and structure of the marine environments. Thus, genetic data works as a basis for management of fish stocks by providing information about the population\'s distribution, structure, and diversity. It is also a highly accurate tool for species identification when diagnosis morphological data are not available. The present work aimed, for the first time, to assess population genetics of Tetrapturus pfluegeri (longbill spearfish) from the Western Atlantic using nuclear and mitochondrial molecular markers. Furthermore, DNA-based forensic tools were applied to find the billfish and swordfish species composition traded in the largest central supply of agricultural products in Latin America, the CEAGESP (General Warehouse Company of São Paulo). Thus, T. pfluegeri showed to be composed by distinct genetic groups with highly significant differentiation (FST = 0,075 - 0,198; p < 0,05) (DEST = 0,092 - 0,176; p < 0,05), probably caused by reproductive philopatry process. From the 432 samples collected in CEAGESP, 78,01% corresponded to Xiphias gladius, 20,83% to Istiophorus platypterus, and 0,23% to Tetrapturus georgii. It provides evidence that molecular tools are highly efficient to overcome the difficulty of billfish identification and the illegal capture of overexploited species in the Atlantic Ocean
