FUS and TDP-43 are two RNA binding proteins sharing similar structures. They are involved in several functions of RNA processing, such as RNA transport, splicing, transcription, as well as in cell compartmentalization. Despite these similarities, they exhibit distinct behaviours in their RNA-binding modes. FUS can bind RNA non-specifically, while TDP-43 is very specific for GU motif. These two proteins have been found aggregated in cytoplasmic inclusions of neurons from patients with overlapped neurodegenerative diseases as ALS, FTLD, and LATE. The fact that these two proteins were co-aggregated in the same inclusions, and are involved in the same cellular complexes, suggests that they are implicated in common pathways.
