(A) dsRNA was purified from two individual virus preparations using either GenElute Mammalian Total RNA Miniprep kit (RTN350, Millipore Sigma), or by conventional RNA precipitation. RNA products were resolved by agarose gel electrophoresis and visualized under UV light. (B-C) L929 cells were infected with reovirus at an MOI of 3 with or without cycloheximide (100μg/mL) and samples were collected every 3 hours, purified through Mammalian Total RNA Miniprep kit, subjected to RT with random primers and qPCR with specific primers to measure RNA levels for each reovirus gene.
