Effect of vitamin D3 on hormonal and reproductive parameters of rats with induced polycystic ovary syndrome.

Zespół policystycznych jajników (PCOS) jest jednym z najczęściej występujących zaburzeń hormonalnych u kobiet w wieku rozrodczym i może dotyczyć nawet 21% kobiet. Charakteryzuje się zaburzeniami reprodukcyjnymi, hormonalnymi i metabolicznymi, tj. zaburzenia/zatrzymanie owulacji, cysty jajników w obrazie USG, hiperandrogenizm, hiperinsulinemia czy insulinooporność. Coraz więcej doniesień wskazuje, że witamina D3 może być skuteczną alternatywą w leczeniu PCOS. Ponadto zaobserwowano jej niedobór u wielu kobiet cierpiących na to schorzenie. Celem niniejszej pracy było zbadanie wpływu suplementacji witaminy D3 na parametry hormonalne i reprodukcyjne u szczurzyc z indukowanym PCOS. W tym celu, 32 samice szczurów rasy Wistar przydzielono losowo do czterech grup: kontrolnej (K; n=8), suplementowanej witaminą D3 w dawce 500IU na dzień (VD; n=8), z indukowanym PCOS za pomocą letrozolu w dawce 1 mg/kg masy ciała na dzień (L; n=8) oraz grupy z indukowanym PCOS, którym podawano jednocześnie witaminę D3 (VD+L; n=8). Przez 21 dni doświadczenia szczury codziennie ważono oraz monitorowano przebieg cyklu płciowego poprzez analizę mikroskopową wymazów z pochwy. Po uśmierceniu szczurzyc, pobrano krew w celu analizy stężenia estradiolu i testosteronu za pomocą testu immunoenzymatycznego ELISA oraz jajniki, które utrwalono w celu zweryfikowania rozwoju PCOS. Analiza morfologiczna jajników potwierdziła skuteczność zastosowania letrozolu, który jest inhibitorem aromatazy, dając obraz wielu cyst jajnikowych. Ponadto zwierzęta w grupie L miały wysoki poziom testosteronu i niski poziom estradiolu, a także zatrzymał się u nich cykl rujowy. W grupie VD+L, zaobserwowano obniżenie poziomu testosteronu w stosunku do grupy L, jednak poziom estradiolu nie uległ zwiększeniu. W grupach, w których podawano witaminę (VD i VD+L) zanotowano skrócenie cykli oraz zwiększenie ich liczby. Obraz morfologiczny jajników szczurzyc, którym podawano witaminę D3 (VD),charakteryzował się gęstym zrębem jajnika oraz większą ilością pęcherzyków przedantralnych w porównaniu do grupy kontrolnej (K). Obrazy morfologiczne w grupach z indukowanym PCOS (L i VD+L) charakteryzowały się obecnością licznych cyst jajnikowych, jednak w grupie leczonej witaminą (VD+L) stwierdzono obecność pojedynczych pęcherzyków antralnych. Otrzymane wyniki sugerująpozytywny wpływ witaminy D3 na parametry hormonalne i reprodukcyjne w PCOS, co daje nadzieję na jej skuteczne wykorzystanie w terapii tego schorzenia.Polycystic ovary syndrome (PCOS) is one of the most common hormonal disorders in women of reproductive age and it may affect up to 21% of women. It is characterized by reproductive, hormonal and metabolic disorders, i.e. ovulation disorders/arrest, ovarian cysts in the ultrasound, hyperandrogenism, hyperinsulinemia or insulin resistance. More and more reports indicate that vitamin D3 could be an effective alternative therapy in the PCOS treatment. In addition, its deficiency has been observed in many women suffering from this disease. The aim of this study was to investigate the effect of vitamin D3 supplementation on hormonal and reproductive parameters of rats with letrozole-induced PCOS. For this purpose, 32 female Wistar rats were divided into four groups: control (K; n=8), vitamin D3 treated at dose 500IU daily (VD; n=8), withletrozole-induced PCOS at dose 1mg/kg body weight daily (L; n=8) and PCOS-induced treated with vitamin D3 (VD+L; n=8). Rats were weighed daily for 21 consecutive days of the experiment and the course of estrous cycle was monitored by microscopic analysis of vaginal smears. After killing the animals, blood was collected for analysis of estradiol and testosterone concentration using an enzyme-linked immunosorbent assay(ELISA) and ovaries were fixed to verify development of PCOS. Morphological analysis of the ovaries confirmed the effectiveness of the letrozole, which is aromatase inhibitor inducing multiple ovarian cysts. Furthermore, rats in the L group had high testosterone and low estradiol levels and were acyclic. In the VD+L group, we observed a decrease in testosterone level compared to the L group, however estradiol level did not increase. In the groups, which were vitamin D3-treated (VD and VD+L) the cycles were shorter and more frequent. The morphological images of the ovaries of vitamin D3-treated rats were characterized by dense ovarian stroma and a higher number of preantral follicles compared to the control group (K). The morphological images of the PCOS-induced groups (L and VD+L) were characterized by the presence of numerous ovarian cysts. However, in the groups treated with vitamin D3 (VD+L), single antral follicles were observed. The obtained results suggest a positive effect of vitamin D3 on the hormonal and reproductive parameters in PCOS, which gives hope for its effective use in the treatment of PCOS
