Claudin-5 based immunohistochemistry studies in equine haemangiosarcoma

A lovak haemangiosarcomájának claudin-5 alapú immunhisztokémiai vizsgálata során a SZIE-ÁOTK Kórbonctani és Igazságügyi Állatorvostani tanszék 2008-2012 között beérkezett tumorarchívumát és az Üllői Nagyállat Klinika Kórbonctani osztályának diagnosztikai boncolási eseteiből gyűjtött mintákat dolgoztuk fel összehasonlító célzattal. A claudin-5 expressziós immunhisztokémiai vizsgálatok alapján a vascularis endothel eredetű daganatok (haemangioma, haemangiosarcoma), a granulációs szövetek (krónikus bőrlob) illetve az intakt vér és nyirokerek anti-claudin-5 antitesttel pozitív, míg az epithelialis (carcinoma, papilloma) és mesenchymalis (fibrosarcoma) daganatok negatív reakciót adtak minden minta esetében. A claudin-5 fehérje kimutatása immunhisztokémiai módszerrel a hisztopatológiai differenciál diagnózis fontos eszközévé vált a haemangioma és a haemangiosarcoma lovakban történő egyértelmű azonosítására. Azonban annak kiderítésére, hogy milyen szerepet játszik a claudin-5 expressziója a tumor pathológiájában, még további vizsgálatok szükségesek.In this study, we examined the equine haemangiosarcoma with claudin-5 based immunohistochemistry. The equine biopsy és necropsy samples received at the Department of Pathology and Forensic Veterinary Medicine of the Faculty of Veterinary Science, Szent István University, between January 2008 and June 2012, and at the Large Animal Clinic of Üllő. During the immunohistochemical examination of claudin-5 expression we compared the samples with each other. The tumours of vascular endothelial origin (haemangioma, haemangiosarcoma), the intact vessels, and the angiofibroblast tissue (chronic dermatitis) were positives, until the tumours of epithelial (carcinoma, papilloma) and mesenchymal (fibrosarcoma) origin were negatives with claudin-5 antibody. The positive results suggest, that claudin-5 becomes a very important marker to make a difference between haemangioma/haemangiosarcoma and epithelial/mesenchymal tumours, with this protein with an unambiguous way we can identify the tumours of vascular endothelial origin. Further studies are needed to explore the expression of these proteins in vascular tumor pathology
