Microsatellite instability (MSI) is a common tumor phenotype associated with many human cancers. It is a consequence of the deficiency of the MMR (mismatch repair) system. Clinically, MSI cancers have greatly benefited in recent years from the advent of Immune Checkpoint Protein Inhibitors (ICKi), which have led to significant therapeutic advances, particularly in the treatment of patients with metastatic MSI cancer. In the era of immunotherapy, the detection of the MSI phenotype is therefore a major challenge in clinical oncology. Another challenge is to detect the predictive factors of sensitivity or resistance to these new precision therapies in patients.
