Maintenance of telomere length, required for the unlimited cell proliferation displayed by cancer cells, is provided by telomerase, a ribonucleoprotein complex containing a specialized reverse transcriptase, encoded by TERT gene, that uses an internal RNA template to maintain telomeres length, thus playing a critical role in tumor formation and progression. TERT is usually repressed in normal somatic cells, but is detectable in the vast majority of tumors. Recent studies have suggested that TERT, besides maintaining telomere, is involved in other cellular functions, and it may contribute to carcinogenesis also via telomere length-independent mechanisms; therefore its inhibition could represent a promising strategy to improve cancer treatm...
