Production and study of human parathyroid hormone antibodies.

In this study, the immunogenicity of hPTH(1-34) was assessed in mice and rats, using different routes of immunization and two forms of hPTH(1-34), free and conjugated. The responder rats and mice were used for the production of monoclonal antibodies to hPTH(1-34). Several mouse-mouse and rat-mouse hybridoma cell lines were produced. The antibodies secreted were examined for various characteristics and used for the detection of hPTH(1-34) in biological specimens. Polyclonal antibody was also produced in goat against the free hPTH(1-34). Lymphocytes isolated from goat blood were thereafter used in the production of monoclonal antibodies. Fusion of goat B-lymphocytes with both mouse NS1 myeloma cell line and heteromyeloma (ovine-murine hybridoma) resulted in five cell lines secreting goat monoclonal antibodies to hPTH(1-34)
