The stimulation of heart glycolysis by insulin is due to an increase in glucose transport and to activation of 6-phosphofructo-2-linase (PFK-2), the enzyme responsible for fructose 2,6-biphosphate (Fr u-2 ,6-P2) synthesis. Fru-2,6-P2 is a potent stimulator of 6-phosphofructo-1-kinase, a key enzyme of glycolysis. The activation of PFK-2 by insulin is sensitive to wortmannin, a phosphatidylinositol 3-kinase (PI 3-kinase) inhibitor, but is not blocked by rapamycin, which prevents p70 ribosomal S6 kinase activation. A good candidate for mediating the insulin-induced activation of PFK-2 is therefore protein-kinase B (PKB), which is located downstream of PI 3-kinase in the insulin signalling pathways.
