A genetically engineered E.coli strain, ST10-46, has been shown to carry the hemD locus, encoding uroporphyrinogen III synthase (cosynthase) using biochemical and genetic techniques. The strain was produced originally by subcloning a 2.89kb Sau3A fragment from E.coli K-12 chromosomal DNA, maintained within the Carbon and Clarke plasmid pLC41-4, into the BamHI site of pBR322. Direct assay of sonicated extracts of strain ST1046 showed the level of uroporphyrinogen III synthase activity to be 200 times higher than that of the wild type control.
