동맥경화와 유방암 발생에서 CAP1의 역할 연구

Adenylyl cyclase-associated protein 1 (CAP1) as a novel functional receptor for human resistin and clarified its intracellular signaling pathway to modulate inflammatory action of monocytes. I noticed that globular C-terminus of Resistin is structurally similar to the C-terminal cysteine-rich domain (CRD) of PCSK9. Proprotein convertase subtilisin/kexin type-9 (PCSK9) has emerged as a promising therapeutic target for atherosclerotic cardiovascular diseases. However, it has not been elucidated whether PCSK9 mediates inflammation independent to lipid changes, thereby directly effects on the atherosclerotic plaque development. Recently, we identified adenylyl cyclase-associated protein 1 (CAP1) interacts with PCSK9 and regulates PCSK9-mediated lysosomal degradation of LDLR and LDL-C levels. PCSK9 activates NF-kB and turns on the pro-inflammatory cytokines, including TNFα, IL-1β, and IL-6 in the monocytes. Overexpression of PCSK9 in LDLR-/- mice using adeno-associated virus aggravated atherosclerotic plaque development implying the inflammatory ability of PCSK9 beyond LDL-C lowering effect. Syk and PKCδ were identified for the CAP1 interacting proteins by advanced liquid chromatography-tandem mass spectrometry (LC-MS/MS). CAP1 deficient mice developed less atherosclerotic lesions, and showed less macrophage accumulation and lower mRNA level of pro-inflammatory molecules (TNFα, IL-1β, IL-6) than the control mice. To further establish role of CAP1 in cancer, I made TALEN-mediated CAP1 knock-out MDA-MB 231. CAP1 deletion MDA-MB-231 cell shows remarkably reduced cell growth and migration under treatment of epidermal growth factor (EGF) in vitro. CAP1 deletion cells cannot progress cell cycle effectively due to G1 phase arrest and decrease actin filament dynamics. CAP1 deletion MDA-MB-231 reduced activation of Src protein for regulating actin filament. CAP1 interacted with Src and is related to locating Src to plasma membrane that is essential for its activation. Inhibition peptide for interaction between CAP1 and Src is designed, and this peptide effectively reduces activation of Src in MDA-MB-231. In conclusion, PCSK9-induced inflammation was mediated by PKCδ and Syk phosphorylation, and its mechanism was dependent on binding between CAP1 and PCSK9. Activation of Src was dependent on binding of CAP1 and inhibition of CAP1 and Src interaction can lead cancer progression and metastasis.Resistin은 단핵구를 자극하고 혈관 염증을 유도하여 동맥 경화증을 악화시키는 아디포카인 입니다. Adenylyl cyclase-related protein 1 (CAP1)은 레지스틴에 대한 새로운 기능적 수용체로서 단핵구의 염증 작용을 조절하는 세포 내 신호 전달 경로를 명확히 했습니다. Resistin의 구형 C- 말단은 PCSK9의 C- 말단 시스테인이 풍부한 도메인 (CRD)과 구조적으로 유사하다는 것을 알았습니다. Proprotein convertase subtilisin / kexin type-9 (PCSK9)는 동맥경화성 심혈관 질환에 대한 유망한 치료 표적으로 부상했습니다. 그러나 PCSK9이 지질 변화와 무관하게 염증을 매개하여 동맥 경화성 플라크 발생에 직접적인 영향을 미치는지 여부는 구체적으로 밝혀지지 않았습니다. 최근에 CAP1이 PCSK9와 상호 작용하고 LDLR 및 LDL-C 수준의 PCSK9 매개 리소좀 분해를 조절한다는 것을 확인했습니다. PCSK9는 NF-kB를 활성화하고 단핵구에서 TNFα, IL-1β 및 IL-6을 포함한 염증성 사이토 카인의 발현을 증가시켰습니다. LDLR-/-마우스에서 아데노 관련 바이러스를 사용한 PCSK9의 과발현은 LDL-C 저하 효과를 넘어서는 PCSK9의 염증 능력을 암시하는 동맥 경화성 플라크 발달을 악화시켰습니다. 고급 액체 크로마토 그래피-탠덤 질량 분석법 (LC-MS / MS)에 의해 Syk 및 PKCδ가 CAP1과 상호 작용 단백질로 확인되었습니다. CAP1이 결핍 된 마우스는 동백경화성 병변이 덜 발생하고 대조군 마우스보다 대식세포 축적이 적고 전 염증 분자 (TNFα, IL-1β, IL-6)의 mRNA 수준이 더 낮았습니다. 암에서 CAP1의 역할을 더욱 확립하기 위해 TALEN 매개 CAP1 녹아웃 MDA-MB 231을 만들었습니다. CAP1 결실 MDA-MB-231 세포는, 표피 성장 인자 (EGF) 처리 조건에서 세포 성장과 이동이 현저하게 감소 된 것을 보여줍니다. CAP1 결실 세포는 G1 단계 정지로 인해 세포주기를 효과적으로 진행할 수 없으며 액틴 필라멘트 역학이 감소하였습니다. CAP1 결실 MDA-MB-231은 액틴 필라멘트 조절을 위한 Src 단백질의 활성화를 감소시켰습니다. CAP1은 Src와 상호 작용했으며 Src를 활성화에 필수적인 세포막에 위치시키는 것과 관련이 있습니다. CAP1과 Src 간의 상호 작용을 억제하기 위한 펩타이드를 제작했으며, 이 펩타이드는 MDA-MB-231에서 Src활성화를 효과적으로 감소시켰습니다. 결론적으로, PCSK9에 의한 염증반응은 Syk 와 PKCδ인산화에 의해 매개되었고 그 메커니즘은 CAP1과 PCSK9 사이의 결합에 의존적이었다.
