Valorization of extracts of medicinal plants endemic to the Middle East : chemical characterization and biological properties (inhibition of enzymes, cancer cells and bacterial strains)

Malgré sa superficie relativement réduite (10452 Km2), la flore végétale libanaise possède un très haut niveau de biodiversité et un taux d’endémisme élevé. Au sein du bassin méditerranéen qui est riche en diversification, la région libanaise est considérée comme un «point chaud régional» à part entière. Vu que les recherches scientifiques sur les molécules d'intérêt des plantes Libanaises ont été focalisées sur les composés volatiles (huiles essentielles), les molécules non-volatiles sont très peu exploitées et rarement publiées. Par la suite, l’intérêt de cette étude a été de réaliser un criblage phytochimique et biologique des extraits de sept espèces de plantes endémiques: Stachys ehrenbergii Boiss., Stachys nivea Labill, Cousinia libanotica D.C., Berberis libanotica C. K. Shneider, Origanum ehrenbergii Boiss., Origanum syriacum L. et Salvia fruticosa Miller. Une extraction par macération à froid avec des solvants à polarité croissante a été réalisée (cyclohexane, dichlorométhane, acétate d’éthyle et méthanol). La famille des polyphénols totaux a été dosée. Il s’est avéré qu’elle contribue majoritairement au pouvoir antioxidant concrétisé par la mesure de la capacité des extraits a inhibé le radical DPPH. L’identification des molécules des extraits a été effectuée selon les analyses HPLC-DAD et GC-MS. Des molécules phénoliques, des dérivés méthoxyphénols, des stérols, des flavonoïdes et des acides gras ont été confirmés. Les activités anti-Alzheimer (anti-acétylcholinestérase), anti-xanthine oxydase (anti-XOD), anti-inflammatoire (anti-15 lipooxygénase), cytotoxique (inhibition de la prolifération des lignées cellulaires HCT-116 ; MCF-7 et Caco-2) et antibactérienne ont été testées et uniquement celles qui ont montré des résultats significatifs ont été listées. L’extrait O. ehrenbergii-Dcm a montré une activité antibactérienne importante contre les souches de Listeria monocytogenes (MIC= 4-10 μg/mL). L’extrait S. fruticosa-Dcm a été responsable d’une activité cytotoxique significative contre les lignées de Caco-2 (CI50= 24.0 μg/mL) et HCT-116 (CI50= 19.7 μg/mL). L'extrait cyclohexane de C. libanotica a inhibé la prolifération de la lignée cellulaire cancéreuse Caco-2 de 46.5%. L’organe fruit de la partie aérienne de B. libanotica extrait par le Dcm a inhibé l’enzyme AChE de 65.3% à 50 μg/mL. A cette même concentration, l’inhibition de l’AChE avec les extraits de S. ehrenbergii et S. nivea a varié entre 52 et 62%. L’extrait méthanolique d’O. syriacum a réussi à piéger 49.6% du DPPH.Despite its relatively small area (10.452 km2), the Lebanese flora has a very high level of biodiversity and a high rate of endemism. Within the Mediterranean basin which is rich in diversification, the Lebanese region is considered as a regional hotspot. Scientific research on the molecules of interest in Lebanese plants has been focused on volatile compounds of essential oils. The non-volatile molecules are very little investigated and rarely published. Hence, the aim of this study was to perform a phytochemical and biological screening of the extracts of seven endemic plant species: Stachys ehrenbergii Boiss., Stachys nivea Labill, Cousinia libanotica D.C., Berberis libanotica C.K. Shneider, Origanum ehrenbergii Boiss., Origanum syriacum L. and Salvia fruticosa Miller. Cold maceration extraction with solvents presenting an increasing polarity was performed (cyclohexane, dichloromethane, ethyl acetate and methanol). The total polyphenol content was assessed and was shown to greatly contribute to the antioxidant activity measured by the capacity of the extracts to inhibit the DPPH radical. The identification of the molecules was carried out using the HPLC-DAD and GC-MS analyses. Phenolic molecules, methoxyphenol derivatives, sterols, flavonoids and fatty acids were identified. Anti-Alzheimer (anti-acetylcholinesterase), anti-xanthine oxidase (anti-XOD), anti-inflammatory (anti-15-lipooxygenase), cytotoxicity (growth inhibition of MCF-7; HCT-116 and Caco-2 cells) and antibacterial activities were tested and only the ones that gave significant results were presented. The O. ehrenbergii-Dcm extract showed an important antibacterial activity against the strains of Listeria monocytogenes (MIC = 4-10 μg/mL). The extract of S. fruticosa-Dcm had a significant cytotoxic activity against the Caco-2 cells (IC50 = 24.0 μg/mL) and the HCT-116 cells (IC50 = 19.7 μg/mL). The cyclohexane extract of C. libanotica inhibited the proliferation of the cancer cell line Caco-2 by 46.5%. The fruit part of B. libanotica extracted by Dcm inhibited the acetylcholinesterase by 65.3% at 50 μg/mL.
