From ancient time, humanity uses natural products for healing purposes. The first use was made from the ingestion of parts of plants such as leaves and fruits. These therapeutic characteristics come from the secondary metabolites that can be found in plant species, animal, and microorganism. Among microorganisms, endophytic fungi have been widely studied because of the variety of bioactive chemicals. In this work, we carried out an optimization in obtaining organic extracts from the culture medium and mycelial mass of the endophytic fungus B4-3L, isolated from the Humirianthera ampla, and we performed tests against the cytotoxic and antibacterial activity. We incubated the strain B4-3L in semi-solid culture medium Potato Dextrose Agar (BDA) for seven days, static at 25 °C. After that, we inoculated the fungus into four different types of culture media, including three liquid media: Potato-Dextrose (BD), Meat Extract and Peptone (MEP) and Sabouraud-Dextrose (SBD), and the solid rice culture. We used four extraction methods to obtain the organic extracts: maceration (M), ultrasound (U), ultrasound associated with maceration (U+M) and liquid-liquid partition (ML). From statistical analyzes, we identified that the best conditions to obtain a more significant amount of organic extract mass were using the SBD culture medium for fungal growth and extraction by the maceration method. With the analysis of the chromatographic profile, it was possible to observe a change in the production of compounds in the organic extracts obtained in the different growth media used. The results of the cytotoxic and antibacterial activities showed that the extracts obtained by growth in SBD and BD, extracted by the liquidliquid partition method, were the most active, since they had a percentage of inhibition >80% in the three strains tested (colorectal, glioblastoma and prostate), also presenting an inhibitory profile against the bacterium Staphylococcus aureus.Desde os tempos antigos a humanidade usa produtos naturais para fins curativos, essa primeira utilização foi feita a partir da ingestão de partes de plantas como as folhas e os frutos. Essas características terapêuticas advêm dos metabólitos secundários que podem ser encontrados em espécies de plantas, animais e microrganismos. Dentre os microrganismos, os fungos endofíticos vêm sendo muito estudados devido à variedade de substâncias químicas bioativas. Com isso, esse trabalho realizou uma otimização na obtenção de extratos orgânicos a partir do meio de cultivo e massa micelial do fungo endofítico B4-3L isolado da planta Humirianthera ampla e realizou testes frente a atividade citotóxica e antibacteriana. A cepa B4-3L foi incubada em meio de cultura semissólido Batata Dextrose Agar (BDA) por 7 dias, estático a 25 ºC. Após o processo de incubação o fungo foi inoculado em quatro tipos diferentes de meios de cultura, sendo três meios líquidos: Batata-Dextrose (BD), Extrato de Carne e Peptona (MEP) e Sabouraud-Dextrose (SBD) e o meio de cultura sólido de Arroz. Foram utilizados quatro métodos de extração para obtenção dos extratos orgânicos: maceração (M), ultrassom (U), ultrassom associado à maceração (U+M) e partição líquido-líquido (ML). A partir de análises estatísticas foi identificado que as melhores condições para se obter uma maior quantidade de massa de extrato orgânico foi utilizando o meio de cultura SBD para o crescimento fúngico e extração pelo método de maceração. Com a análise do perfil cromatográfico foi possível observar uma mudança na produção de compostos nos extratos orgânicos obtidos nos diferentes meios de crescimento utilizados. Os resultados das atividades citotóxica e antibacteriana mostraram que os extratos obtidos pelo crescimento em SBD e BD extraídos pelo método de partição líquido-líquido apresentaram-se como os mais ativos, pois tiveram percentual de inibição >80% nas três linhagens testadas (colorretal, glioblastoma e da próstata), apresentando também um perfil inibitório frente a bactéria Staphytlococcus aureus.Lagarto, S
