Understanding planetary formation is a major challenge of modern astronomy. It is now established that planetary systems are born within protoplanetary disks, which are formed concomitantly with pre-main sequence stars.This thesis presents the work that I conducted to determine the physicochemical conditions prevailing in these objects. I present especially millimeter interferometry observations of disks surrounding the young stars DM Tau, LkCa 15, MWC 480, AB Aur and HD 34282 in the rotation lines of CO isotopes, and a few radicals and molecules (HCN, CN, etc.). I present also the parametric model that I used to analyze these data, including the non local thermodynamic equilibrium radiative transfer part that I developed during my thesis. ...
