Vitrification of equine ovarian tissue in different resveratrol concentrations: 10 µM and 20 µM

The present study was carried out to compare the different antioxidant solutions in vitrification of equine ovarian tissue, a faster cryopreservation technique compared to slow freezing, evaluating the morphology and viability of vitrified follicles included in the equine ovarian tissue. For that, ovary fragments measuring 3mm x 3mm x 1mm were vitrified on French vane, using two different solutions of vitrification (SV), containing 10 um of resveratrol and 20 um of resveratrol. After 15 days, the fragments of the ovarian tissue were heated and processed for histology for morphological analysis of the follicles. The histological analysis showed that there was no statistical difference between the treatments with SV1Res10 and SV1Res20 in the percentage of morphologically normal follicles after vitrification, with a considerable cytoplasmic retraction of the follicles, and that the control solution was better. In conclusion, vitrification containing resveratrol as an antioxidant for both 10æm and 20æm of resveratrol, so that the time of exposure to the antioxidant agent and temperature, are not most suitable for vitrification of equine ovarian tissue.Trabalho de Conclusão de Curso (Graduação)O vigente estudo foi realizado com a finalidade de comparar as diferentes soluções de antioxidante na vitrificação de tecido ovariano equino, uma técnica de criopreservação mais rápida comparada ao congelamento lento, avaliando a morfologia e viabilidade dos folículos vitrificados inclusos no tecido ovariano equino. Para tanto, fragmentos de ovário medindo 3mm x 3mm x 1mm foram vitrificados em palhetas francesas, utilizando duas diferentes soluções de vitrificação (SV), contendo 10 um de resveratrol e 20 um de resveratrol. Após quinze dias, os fragmentos do tecido ovariano foram aquecidos e processados para histologia para análise morfológica dos folículos. A análise histológica mostrou que não houve diferença estatística entre os tratamentos com SV1Res10 e SV1Res20 no percentual de folículos morfologicamente normais após vitrificação, ocorrendo uma considerável retração citoplasmática dos folículos, e que a solução controle, mostrou­ se melhor. Em conclusão, a Vitrificação contendo resveratrol como antioxidante nas concentrações 10 um e 20 um de resveratrol, não foram suficientes para se obter resultados positivos em relação à normalidade dos folículos ovarianos de equinos e integridade destes nos fragmentos, de modo que o tempo de exposição ao agente antioxidante e a temperatura podem não são a mais adequadas para vitrificação de tecido ovariano equino
