This work is a study of active enzymes by several scanning probe microscopies in order to extract mechanical, electrochemical and electrical properties. It is divided into three parts. Atomic Force Microscopy (AFM) studies of yeast 3-phosphoglycerate kinase (PGK) ex situ and in situ is coupled to UV-visible spectrophotometry to compare the enzyme activity of the immobilized enzyme to that of the free enzyme. Glucose Oxidase and the decomposition of hydrogen peroxide are studied by Scanning Electrochemical Microscopy (SECM) to extract heterogeneous rate constant.
