Antimicrobial effect of Origanum vulgare essential oil on the growth of Listeria monocytogenes with potential use in antimicrobial films applied in the dairy industry.

El objetivo del presente trabajo de investigación fue evaluar el efecto antimicrobiano del aceite esencial (AE) de Orégano Origanum vulgare frente al crecimiento de Listeria monocytogenes, se evaluaron 13 concentraciones del aceite bajo tres métodos para determinar el efecto antimicrobiano del AE, gota sobre césped, método de Kirby Bauer (MKB) y siembra en pozo (MSP); se empleó como control positivo una cepa de Carnobacterium maltaromaticum y como control negativo aceite mineral, los ensayos se realizaron por triplicado, se tomaron las medidas correspondientes de los halos de inhibición con la ayuda de un calibrador de marca Mitutoyo, para obtener medidas más precisas, se promediaron las tres medidas y se determinó el porcentaje de inhibición para cada concentración de aceite esencial. Posterior al tiempo de incubación se descartó el método de siembra en pozo debido que se presentó solapamiento de los halos impidiendo tomar las respectivas medidas, de manera que los dos métodos que se tuvieron en cuenta fueron MKB y MSP. A partir de los dos métodos de siembra se obtuvo un efecto antimicrobiano del AE de orégano frente a L. monocytogenes, en todas las concentraciones estudiadas se logró obtener halos de inhibición hasta la concentración 11 que iban desde los 3,7 cm hasta los 0,1 cm. Las dos concentraciones con comportamientos antimicrobianos estables fueron la 33 y 25, las cuales permitieron obtener porcentajes de inhibición de 32,2% a 39,6%, lo que podría indicar una concentración de aceite esencial adecuada para su potencial uso en películas antimicrobianas aplicadas en la industria láctea.The objective of this research was to evaluate the antimicrobial effect of the essential oil (EO) of Origanum vulgare against the growth of Listeria monocytogenes, 13 concentrations of the oil were evaluated under three methods to determine the antimicrobial effect of the EO, drop on grass, Kirby Bauer method (MKB) and sowing in well (MSP); A strain of Carnobacterium maltaromaticum was used as a positive control and mineral oil as a negative control; the tests were performed in triplicate, the corresponding measurements of the inhibition halos were taken with the help of a Mitutoyo calibrator to obtain more precise measurements, the three measurements were averaged and the percentage of inhibition was determined for each concentration of essential oil. After the incubation time, the well sowing method was discarded because of the overlapping of the halos that prevented taking the respective measurements, so the two methods that were taken into account were MKB and MSP. From the two seeding methods, an antimicrobial effect of oregano EO against L. monocytogenes was obtained, in all the concentrations studied, inhibition halos were obtained up to concentration 11, ranging from 3.7 cm to 0.1 cm. The two concentrations with stable antimicrobial behavior were 33 and 25, which allowed obtaining inhibition percentages of 32.2% to 39.6%, which could indicate a concentration of essential oil suitable for potential use in antimicrobial films applied in the dairy industry.Microbiólogo (a) IndustrialPregrad
